Purpose:To observe the effects of Huangqi Gegen decoction and itscompatibility on liver lipid metabolism in diabetic rats.We aimed toanalyze the Astragaliradix and Puerariaelobataeradix in the liverglycolipid metabolism in diabetic rats.Methods:86SD male rats were randomly divided into6group,normal group、model grou、 positive control group、 Astragaliradix group、Puerariaelobataeradix group and Huangqi Gegen decoction group,14eachgroup,except normol group(11rats).In addition to the normal group andpositive control group,the rest of the group were induced by STZ(46mg·kg-1)made after the model according to the2×2factorial designexperiment scheme.Positive control group by1.47g·kg-1body weight dosegavage, Astragaliradix group, Puerariaelobataeradix group, Huangqi Gegendecoction group were2.7g·kg-1,1.35g·kg-1,4.05g·kg-1body weight doseirrigation stomach.After STZ treatment(30days),blood glucose level ofall rats were measured in group2-6.In serum,CHO and TG were tested by autobiochemistry analyzer.We assayed ADPN、GLUT4、INS、the impact of leptin、IL-12and IL-15in liver tissue by ELISA.Liver’s Adipo R2and PPARαwere analyzed by RT-PCR.We also assayed protein level Adipo R2and PPARαin liver by western blotting.The dates analyzed by SPSS software.Results:1Effect on pharmacodynamics index of Huangqi Gegen decoction group indiabetic rat. 1.1Effect on blood glucoseIt’s no statistic difference in blood glucose among the rats inGroup1-6. After STZ ip,blood sugar in the STZ group elevated than in normalgroup(P<0.05).The30th days after STZ treatment,blood glucosesignificantly increased in the model group comparing with the normal one,while blood sugar in the others were more decreased than model group(P<0.05).1.2Effect on blood lipidIn model group,CHO and TG in serum were more elevated than normalgroup, it has meaning on statistics(P<0.05).Compared with serum CHO inmodel group with other groups,CHO were decreased remarkably inAstragaliradix,Puerariaelobataeradix and Huangqi Gegen decoction group(P<0.05).In treatment groups(except Huangqi Gegen decoction group),TGlevel were clearly decreased in serum (P<0.05).Compared withastragaliradix and Puerariaelobataeradix:(1)Effect on single index ofserum CHO or TG: It’s definitely different from Astragaliradix group、Puerariaelobataeradix group and model group,but astragaliradix andpuerariaelobataeradix had the same effects.(2)Effect on comprehensiveindex of serum CHO or TG: It’s remarkably different from Astragaliradixgroup、Puerariaelobataeradix group and model group,but astragaliradixand puerariaelobataeradix had the same effects.Analyzed the interactionwith astragaliradix and puerariaelobataeradix:astragaliradix andpuerariaelobataeradix have non-interaction to serum CHO, but they hadinteraction to serum TG, compability with each other worked better thansingle one.2Huangqi Gegen decoction diabetic rat liver ADPN、GLUT4、 INS, the impactof leptinRat liver model group ADPN、GLUT4、INS,leptin were significantlydecreased compared with normal group differences were statistically significant(P<0.05).Between single herbal medicine group:(1)the impactof ADPN: Astragaliradix,Puerariaelobataeradix group and model group werenot sig-nifycantly different;(2) GLUT4impact: Astragaliradix group andmodel group were significantly different,Puerariaelobataeradix ’s notobvious;(3) the impact on INS: Astragaliradix group and model group weresigni-ficantly different,Puerariaelobataeradix is not obvious;(4) onleptin effects:Astragaliradix,Puerariaelobataeradix and model groupwere sig-nificantly different effects of Astragaliradix and Puerariae-lobataeradix different,Astragaliradix role better. Between the maineffects and drug interactions analysis:(1) effect on ADPN: Astragaliradix,Puerariaelo-bataeradix had no effect on ADPN, there is no interactionbetween the two drugs;(2)effect on GLUT4: Astragaliradix can effectivelyenhance GLUT4levels,Puerariaelobataeradix is not obvious,there was nosignificant interaction between the two drugs;(3) the role of INS:INSAstragaliradix can effectively improve insulin levels, Puerariaeloba-taeradix is not obvious, no significant interaction between the two drugsmeaning;(4)the role of leptin: Astragaliradix,Puerariaelobataeradix caneffectively enhance leptin levels,there are interactions between the twodrugs,the combination is better than single drug effects.3Effect· on IL-12and IL-15in liver tissueIt has statistic significance to the difference compared normal groupand model groups which IL-12and IL-15in liver tissue were increasedobvious-sly(P<0.05).Compared with IL-12level of model group with theother groups,IL-12level were decreased remarkably in Astragaliradix,Puera-riaelobataeradix and Huangqi Gegen decoction group(P<0.05);IL-15level in each treatment group clearly declined (P<0.05).Compared withastra-galiradix and Puerariaelobataeradix:(1) Effect on IL-12:Thedefinite difference from Astragaliradix group、Puerariaelobataeradixgroup and model group had statistic significance, while astragaliradix and puera-riaelobataeradix had the same effects.(2) Effect on IL-15:It’s definitely different from Astragaliradix group、Puerariaelobataeradixgroup and model group, had statistic significance, astragaliradix workedbetter than puerariaelobataeradix,so their effects were different.(3)Effect on comprehensive index of IL-12and IL-15: It’s remarkablydifferent from Astragaliradix group、Puerariaelobataeradix group.Theyhave different influence,but Astragaliradix group worked better.Analyzedthe intera-ction with astragaliradix and puerariaelobataeradix:(1)Effect on IL-12: astragaliradix could decrease IL-12level,but puerariae-lobataeradix couldn’t.They interacted with each other,but compabilitywith each worked worse than astragaliradix singly.(2)Effect on IL-15:Both astra-galiradix and puerariaelobataeradix could decrease IL-15level.They interacted with each other,but compability of each workedbetter than each one.Effect on comprehensive index of IL-12and IL-15:Bothastragaliradix and puerariaelobataeradix had effect on comprehensiveindex of IL-12and IL-15.They interacted with each other,but compabilityof each worked better than each one.4Effect on Adipo R2and PPARα in liver protein level expressionCompared with the normal group,model group Adipo R2,PPARαmRNAexpression levels were significantly lower, with statisticalsignificance(P<0.01); compared with the model group, liver Huangqi Gegendecoction group Adipo R2mRNA expression level increases,there isstatistically significant(P<0.01);Astragaliradix group, Puerariae-lobataeradix group, Astragaliradix rat liver PPARαmRNA expression ofPuerariaelobataeradix group average increase was statistically signi-ficant(P<0.05).Compared with the control group of traditional Chinesemedicine, Puerariaelobataeradix group,Huangqi Gegen decoction groupAdipo R2groups can increase gene expression levels(P<0.05);upregulatedPPARαmRNA expression in each treatment group was not statistically different.Compared with the normal group,the rat liver model group AdipoR2and PPARα protein levels were significantly decreased,with statis-tical significance(P<0.05). Compared with the model group, the Astra-galiradix group, Puerariaelobataeradix group,elevated liver HuangqiGegen decoction group Adipo R2,PPARαprotein expression levels werestatistically significant(P<0.05).Compared with the control group oftraditional Chinese medicine,Puerariaelobataeradix group,rat liverHuangqi Gegen decoction group Adipo R2protein levels increase(P<0.05);in protein expression PPARα,no significant difference in each treatmentgroup level.Conclusions:1.Puerariaelobataeradix reduces blood glucose levels in diabetic rats wassuperior to Astragaliradix, did not show efficacy of superimposed, andno interaction.2.Astragaliradix, Puerariaelobataeradix serum triglyceride, cholesterollowering effect of strength of diabetic rats, there exist interactionsin the lower serum triglyceride process, combination is superior to singledrug.3.Astragaliradix decreased in liver tissue of rats with diabetic mellitusleptin,interleukin15levels of function is better than that ofPuerariaelobataeradix, and has interaction, combination is superior tosingle drug.4.Astragaliradix,Puerariaelobataeradix reduce blood sugar,lipids levelsin diabetic rats, and its mechanism may be related with the regulationof adiponectin/Adipo R2/PPARαsignal transduction.
