| Objective:By observing impact of the Aucub to neural function defect scale of cerebral hemorrhage rat and TNF-a expression and white blood cells around the hematoma, investigating whether aucubin by inhibiting TNF-a expression reduce the inflammatory response after cerebral hemorrhage, to play neuroprotective mechanism, to explore a new way for the treatment of cerebral hemorrhage.Methods:90adult male Sprague Dawley rats were randomly divided into sham operation group,cerebral hemorrhage group and aucubin group.Autologous blood was injected into the rat caudate nucleus to prepare cerebral hemorrhage,and sham operation group injected with saline at the same site.Two hours after the surgery,the aucubin group was injected intraperitoneal with aucubin4.2mg/kg once a day,each rat of the sham operation and cerebral hemorrhage rats was injected the same amount of saline.After surgery,each group was randomly divided into five time points,Which were12hã€2dã€3dã€5dã€7d, and at each time point,six rats were teken out until the corresponding point of time.The neural function score of a model rat is evaluated by longa score.The number of white blood cells around the hematoma was measured by HE staining,and the expression of TNF-aaround the hematoma was tested by immunohistochemistry.Result:1.Each time point of cerebral hemorrhage group and aucubin showed obvious symptoms of neurological impairment, neural functional defect appears first increased and then decreased, and reached the peak at3d after operation in all, and neural function improved significantly after Aucubin intervention (P<0.05)。2.HE staining showed that at low magnification, the sham operation group only a little white cell infiltration, and the comparison Of same group at different time points is not statistically significanto (P>0.05) Cerebral hemorrhage group and aucubin group display obvious infiltration of leukocyte12hours after operation, reached the peak at3D, and7d was significantly reduced compared with the peak, But there are still more amount of leucocyte infiltration. The aucubin group is less than the number of leukocyte infiltration in cerebral hemorrhage group (P<0.05)3.Immunohistochemistry showed that sham operation group only have a few deep brown positive staining cells, and the comparison Of same group at different time points is not statistically significant (P>0.05)。Brain hematoma region of cerebral hemorrhage group and cerebral hematoma aucubin group appears dark brown positive staining cells12h after the operation, The two groups of TNF-aimmune response reached the peak3d after the operation。Aucubin group the number of positive cells less than the group of cerebral hemorrhage(P<0.05)。Conclusion:The aucubin has an evident protective effection for injured neuron after cerebral hemorrhage in rats. |
