| Objective:To investigate the effects of total flavone of Clerodendron bungei Steud on liver cancer and possible mechanisms, so as to provide experimental basis for effective anti hepatoma drugs and therapeutic targets.Methods:In vitro culture of HepG2cells, to MTT assay was used to detect different concentrations of clerodendrum bungei total flavonoids in HepG2cells after48h of cell proliferation; by inverted microscope observation of the effects of different concentrations of clerodendrum bungei total flavonoids in HepG2cells after48h of cell morphological changes; influence of flow cytometry smelly peony flavones on cell apoptosis; RT-PCR detection of Clerodendrum flavonoids in HepG2cells after48h of beta catenin and TCF-4mRNA and CD44v6mRNA expression of influence.The inhibitory effects of total flavonoids of cellular level and molecular level of different angles of Clerodendron bungei Steud on liver cancer cells, and to explore its mechanism.Results:1.MTT experiment results showed that the drug concentration of clerodendrum bungei total flavonoids in0.025,0.25,2.5,25,250g/ml and the inhibitory rate was5.55%,12.73%,14.84%,62.44%,76.81%respectively, and blank control groups, the difference was statistically significant difference (P<0.05), suggesting that clerodendrum bungei total flavonoids inhibit the proliferation of HepG2cells.2. The cell morphological observation:under inverted microscope observation of visible the concentration of clerodendrum bungei total flavone group changes in cell morphology, part of the cells become smaller circle, reduce the amount of cytoplasm, in the cytoplasm of a lot of transparent particles, cells were single growth state, adherent ability to drop, shedding increased, part of the cells were suspended growth.3. Flow cytometry showed that with increasing doses of clerodendrum bungei total flavonoids and the proportion of apoptotic cells also increases accordingly, prompt smelly peony total flavonoids can promote the apoptosis of HepG2cells.4.PT-PCR results showed that under the effect of different doses of clerodendrum bungei total flavonoids of beta catenin and TCF-4mRNA and CD44v6mRNA expression decreased, compared with the control group with significant difference.Conclusion:Clerodendrum bungei total flavonoids can inhibit the growth and proliferation of HepG2cells, hepatocellular carcinoma HepG2cell morphology and induced its apoptosis, can down regulate the expression of beta catenin and TCF-4mRNA and CD44v6gene, and the effect is concentration dependent. Clerodendrum bungei total flavonoids induced proliferation and apoptosis in HepG2hepatoma cells change might be related to the regulation of Wnt/beta catenin signaling pathway and inhibit the key genes related to. |
