| Objective:The aim of this study was to investigate the effect and mechanism of acupuncture on characteristic of locomotor activity circadian rhythm and contents of 5-HT,NE and EL-1β in insomnia rat model, to speculation the priciple of adjusting the human Sleep-wake cycle based on the"yang sheng chen,yin sheng ming"theory.Methods:(1)When the 30 SD rats act with the light from the darkness steadily, then divided them into three group randomly,that was blank,model and acupun-cture,with animal recording and analyzing system to observe locomotor activity circadian rhythm.(2)Use intraperitoneal injection of PCPA suspension to copy insomnia SD rats model, and observe the characteristic of locomotor activity circadian rhythm in SD rat model of insomnia, measured by the changes of daily spontaneous activity.(3)Acupun cture was applied to "shenmai" (BL62), and "zhaohai" (KI6) for 20 minutes per day, and lasted for 7 days during 8:00-10:00am.(4)The contents of 5-HT,NE and IL-1β were detected by ELISA method.Results:(1)The disorder of locomotor activity circadian rhythm was expressed by the model of insomnia.The change of insomnia model was the increase of daily spontaneous activity.(2)Compared with model group,the reduction of daily sponta-neous activity could be observed in acupuncture group. (3)Compared with blank and acupuncture groups,after acupuncture,the content of 5-HT was decreased,but NE and IL-1β were significantly increased in model group.(4)Compared with blank and model groups,after acupuncture,the content of 5-HT was increased,but NE and IL-1β were reduced at last in acupuncture group.Conclusions;(1)Unusual behavior and disorder of locomotor activity circadian rhythm were showed in insomnia. (2)Acupuncture could significantly reduce the nunber of daily spontaneous activity. (3)The mechanism of why acupuncture Qiao meridian could treat insomnia perhaps is related with increasing the content of 5-HT and decreasing the content of NE and IL-1β. |
