The dried roots of Euphorbia kansui L.(Euphorbiaceae)are known a toxic traditional Chinese herbal medicine."Gan Sui" was recorded as a low grade drug and has been used as a herbal remedy for edema,ascites,urination and defecation elausura cancer,etc.Previous investigations of E.kansui have yielded numerous ingenane-type diterpenes,macrocyclic jatrophane-type diterpenes,euphane/tirucallane-type triterpenes.The root and trunk bark of Pseudolarix kaempferi Gord.(Pinaceae)are known as "Tu Jin Pi" in traditional Chinese medicine."Tu Jin Pi " has been widely used for treatmeat of dermal infections caused by fungi.Previous studies on the chemical constituents of Tu Jin Pi have yielded numerous diterpenes and phenolic acids.The thesis studies on chemical constituents and bioactivities of E.kansui and P. kaempferi.Sixteen compounds were isolated from 75%alcohol extract of the roots of E.kansui by silica gel,sephadex LH-20 column chromatography,preparative HPLC,etc.Among them, fourteen compounds were elucidated on the basis of physicochemical,sepectroscopic analyses and were characterized as:kansuinin D1(1),kansuinin F1(2),kansuinin A(3), kansuinin B(4),kansuinin D(4),kansuinin F(6),3-O-(2,3-dimethylbutanoyl)-13-O-dodeca-noylingenol(7),20-O-(2,3-dimethylbutanoyl)-13-O-dodecanylingenol(8),3β,20(R)-dihydr-oxydammer-24-methylene(9),betulinic acid(10),kansenone(11),epi-kansenone(12),β-sitosterol(13),isoscopoletin(14).Among them,(1)and(2)were new compounds;(9)and (10)were isolated from this plant for the first time.In addition,the petroleum ether,ethyl acetate and n-butanol fractions from EtOH extract of roots of E.kansui have been screened for in vitro activities.The results revealed that the petroleum ether fraction showed significant activities,which will be helpful to search anti-tumor constituents in the future.Fifteen compounds were isolated from 95%alcohol extract of from the roots of P. kaempferi by silica gel,sephadex LH-20 column chromatography,preparative HPLC,etc. Among them,fourteen compounds were elucidated on the basis of physicochemical, sepectroscopic analyses and were characterized as:ethyl pseudolarate B(17),pseudolaric acid A3(18),methyl pseudolarate B(19),pseudolaric acid A(20)),pseudolaric acid B(21), pseudolaric acid C(22),pseudolaric acid C2(23),deacetylpseudolaric acid A(24), demethoxydeacetoxy pseudolaric acid B(25),pseudolaric acid B O-β-D-glucopyranoside(26), myricetin(27),vaniltic acid(28),gallic acid(29),1,2,3-bezenetriol(30).Among them,(17) and(18)were new compounds;(30)was isolated from this plant for the first time.Pseudolaric acid B(21)has been screened for in vitro activities also.The results displayed that it showed different level in vitro cytotoxic activities against human leukemia HL-60,gastric cancer BGC-823,hepatoma Bel-7402 and nasopharyngeal carcinoma KB,which supply elementary information for further studying anti-tumor activities.
