Expression Of MiRNA In Psoriasis And The Intervention Of Zhuhuang Granule

To confirmed the effect of ZhuHuang granule through clinical observation;To investigate the expression profile of microRNA (miRNA) in psoriasis vulgaris using miRNA array technique,to find out the specific miRNAs is up-regulation or down-regulation, in order to further research it’s roles in the the psoriasis pathogenes; and to detect variation of the expression of these specific miRNAs, to provid new theo-retical and experimental evidence of the action mechanism and targets of using ZhuHuang granule to treat ordinary psoriasis.Methods:30 of the treament group take the ZhuHuang granule,30 of the clinical control group take the tablet Diyin, a month later compare the PASI score.To detect the miRNA expression profile of typical psoriatic plaques and normal person’s skin with miRNA array technique, as well as the leion of the post-treatment psoriatic that have significant curative effect,to find out the changes between the prior and post treatment.Results:Found out 31 specific miRNAs related to psoriasis vulgaris. Comparing to normal persons,10 up-regulated and 21 down-regulated. The 4 cases with significant curative effect,whose prior and post treatment miRNA expression profiles was compared,show that the expression of let-7d*、miR-34a、miR-380*、hsa-miR-185 down-regulated generally,and miR-200c. miR-19b、miR-125b、miR-30c miR-10lup-regulated,other miRNA changed with no regularity.Conclusions:ZhuHuang granule is More effective than tablet Diyin for hot blood type of Psoriasis.MiRNA deregulation is involved in the pathogenesis of psoriasis; ZhuHuang granule may cure the ordinary psoriasis through supressing the specific miRNA’S abnormal expression.
