The resource of goat is one of the most important genetic resource in goat breeding. The development of molecular biology techniques makes it convenient to evaluate the diversity of genetic resources. The objective of this paper is to investigate the genetic diversity of goat by using RAMP markers and to provide valuable information for further utilization of these genetic resources. The main contents of the paper are as follows:1. The effects of concentration of MgCl2, random primers, dNTP, template DNA and Taq DNA polymerase on RAMP were analyzed with goat. An optimalized RAMP-PCR system was set up.2. Genetic diversity of 144 individuals from 11 goat breeds in china was evaluated using RAMP markers. Twenty RAMP primers were used for the PCR amplification of genomic DNA, and 6 primers could produce clear and polymorphic bands. Total 2099 bands were amplified, of which 751(35.7%) were polymorphic. The mean genetic similarity value based on RAMP markers among 144 individuals was 0.401, ranging from0.040 to 0.889. Genetic polarization happened in breeds. The genetic diversity of goat could be evaluated by RAMP markers.3. According to the clustering results based on RAMP markers: all materials of goat could be classified into 4 groups. The results influenced by the environment and artificial selection. Cashmere goat, carnose goat and skinny goat could distinguished by RAMP maker.The above result indicated: RAMP technique could indicate inner breed genetic variations and could provide scientific basis for preservation and cross-breed on indigenous breeds of goat. And it's also a meaningful attempt to apply RAMP technique on goat, that is usually used on plan at present. It had expanded the measures of checking out the genetic variations on animal.
