A molecular genetic linkage map was constructed for an interspecific F1 hybrid population between L. tulipifera Linn(BM)×L. chinense sarg (S) by SSR and SRAP makers. We compared our map with L. chinense sarg (S)×L. tulipifera Linn, the main results were as follows:SRAP makers were applied on the genetic linkage map construction of Liriodendron , we developed the SRAP reaction system of Liriodendron, the amplification of SRAP was in a 25ul volume: 50ng DNA templates, 0.4uM primer of each, 2.0mM Mg2+,0.2mMdNTPs and 0.04units×uL-1 of rTaq enzyme. the results showed that the developed SRAP reaction was stable and reproducible.Joinmap4.0 software was used to construct the genetic map based on the polymorphic loci data tested from the segregating population(BM×S) .Finally , a total of 84 makers were available for mapping on our population(BM×S), including 29 SSRs and 55 SRAPs that distributed in 20 linkage groups. A total 1410.7 cM map length was observed. The average distance of each group was 70.5 cM while the average gap between two makers was 16.8 cM . The estimated genome length was 2201.57 cM in average , the genome coverage of our map was 64%.We firstly compared two linkage maps based on reciprocal cross F1 population. In this experiment, we compared our linkage map with another genetic linkage map based on F1 population (S×BM). The results showed that, 27 homological makers were found from the two genetic maps lied in 5 homologous linkage groups and 8 putative homological linkage groups. The results would provide reference for the research of QTL-mapping and genomic imprinting within Liriodendron.
