We got 10 mononuclear mycelium by collectting spores from pleurotus ostreatus tianda 300 by using spore ejection, dilution,microscopic examination , then mating all the Mononuclear mycelium Respectively .we got 9 dicaryon mycelium Successfully. By using mating tests ,the mating type of the 10 mononuclear mycelium was known . We also found that there is no linkage between their mating type and there phenotypic characteristics .In order to researching into and focused on the difference between mononuclear mycelium and dicaryon mycelium,reasons for the difference and values of the difference, we conducted a series of tests1. Take pleurotus ostreatus tianda 300 cultivated in our laboratory as the material, by using spore ejection, after combined methods as dilution separation,microscopic examination,mycelium growth rate,mycelium thickness and the existence of lock-shaped we got 10 Mononuclear mycelium .2. We got 9 dicaryon mycelium and their mating type by mating all the 10 Mononuclear mycelium.the proportion of the mating type is A1B1:A2B2:A1B2=5:2:3.The phenomenon of segregation distortion of mating-type is exist.We found that there is no correlation between their mating type and theie phenotypic characteristics by comparing the growth rate,level of dense of the mycelium and wether the edge of colony is regular.Also by comparing the growth rate,level of dense of the mycelium and wether the edge of colony is regular,we divided the ten mononuclear mycelium into four types.3.We found their is many differences between the mononuclear mycelium and dicaryon mycelium by analysis spectrum of isozyme of laccase,peroxidase,NADH dehydrogenase,NADPH dehydrogenase,cellulase of mononuclear mycelium 13,3,4,5,6 and dicaryon mycelium 3×3,4×3,13×5,4×5.With the same mobility ratio , mononuclear mycelium and dicaryon mycelium may be have the same isozyme spectrum ,or mononuclear mycelium have isozyme spectrum,but dicaryon mycelium didn't have,or dicaryon mycelium have the isozyme spectrum,but mononuclear mycelium didn't have.4.By comparing the difference between the mononuclear mycelium and dicaryon mycelium we want to probe into the compatibility even fecundity of two mononuclear mycelium.Although this method need to be proved.But it provide backing to the compatibility of pleurotus ostreatus tianda 3005. We analysed and speculated the differences and the reasons of the differences between the isozyme spectrum of mononuclear mycelium and dicaryon mycelium.In order to probe into the mechanization of heterosis.
