Effect Of Mifepristone For Labor Induction Of Term Pregnancy On Kidney Tissue In The Maternal Rats And Their Offsprings

Objective TO investigate the short and long-term influence of mifepristone for laborinduction of term pregnancy on kidney tissue in the maternal rats and their offsprings.Mcthod 20 Wstar rats in 20th gestational day were divided randomly intomifepristone group and control group,while each group wa5 divided short-termgrouP and long-term group.OffSprings were divided according to their mother .Eachrat of mifepristone group was fed on mifepristone ll.5tl.0 mg, while the rat ofcontrol group was fed on sesame oil lmL .In short-term group,the mother rats andtheir neonatal rats was put to death after labor.In long-term group, the mother rats andtheir offsprings were fed to 90 d after postpanum,then to be killed.The ultrastructuralchangs of right kidney were observed by transmission eIectron microscope.Meanwhile, the mitochondria parameters of epitheIial ceIls in proximal tubule anddistal tubule were analysed by stereology in the short-term group.RcsuIt In conparison with control group,the maternal rats kidney tissue ofshort-term mifepristone group displayed:l) renal corPuscle revealed no distinctchanges;2)part of epithelial cells in tubule appeared edematous in cytoplasm; somemitochondria were swollen,their cristae broken;3) interstitium was edematous andloose in texture;4)some interstitial cells were edematous or even showed membranerupture,chromatin margination or condensation.The neonatal rat kidney tissue ofshort-term mifepristone group displayed:l)Some kidney corpuscle endotheliaIcells,podocyte and parietal cel1s were edematous.Their mitochondria were enlargedand cristae were ruPtured. Rough endolasmic reticulum were dilated;2) Most ofepithelia1 cells in proximal tubule appeared edematous; Their mitochondria wereswollen with fragmented cristae,and some even displayed a vacuolardegeneration;3)Parts of epithelial cells in distal tubule and collecting duct wereedematous, Sometimes the nuclei showed chromatin mafgination,Some epithelialcells were seen in the lumen;4) Interstitium was hyperemic and edematous .Someinterstitial cells were edematous or even revealed a ruptured membrane,Their necleiwere condensd or fragmented.In the long-term group,the kidney tissue of maternalrats and their offsprings were not discovered distinct chang morphologically.Thevolum density and average volume of mitochondria of epithelial cells in proxima1and distal tubule of the pregnant rats and their offsprings increased obviously and thenumerical density decreased in the short-term mifepristone grouP.Conclusion Mifepristone fOr labor induction of term pregnancy can cause a damageof kidney in the pregnant rats and their offsprings in the short-term group.
