Radix Astragalus, usually used as a traditional Chinese medicine,has many effects of replenishing "qi" depressing blood pressure stimulating heart dimishing inflammation promoting the subsidence ofswelling,reinforcing immunity and so on.To optimize the extraction and isolation technique of Total Saponinsof Astragalus the orthogonal test was employed to study the effects of theconcetration quantity of ethanol consumed the duration of extractiontime .In the same way,the effects of the type and pH of macroporusresinas well as the concetration of ethanol used as mobile phase in isolationwere :Astragalus was extracted about 90 minutes and 60minutessuccessively in 70% ethanol with a quantity of 8 or 6 times that ofAstragalus using type D21 macroporousresin (pH=7) with a quantity of2.4times that of Astragalus as solicle phase and 90% ethanol as mobitephase.Astragalus was chosen as a model drug ,and was formulated into ofrapidly dissolving tablet in mouth. Order to avoid the disadvantages ofconventional tablet ,designed to dissolve rapidly upon contaction withoral mucous. Rapidly dissolving tablet in mouth without addition of waterconsists of little disintegration agent and sodium sulfate withcrystallization water(Na2SO4·10H2O).??The tablet was prepared by direct compression . The effects ofpreparative variables on the preparation of the tablet were studied by anorthogonal design .Results show that pH and stirring rate of thedissolving media can affect drug release. T50 was proportional to thecontent can of microcrystalline cellulose(MCC) and sodium sulfate(Na2SO4·10H2O) in the carrier while other excipients were remainunchanged.A high perfomance liquid chromatographic method was developedto determine Astragaloside(ASI).Recovery and precision of this methodwere all good .The calibration curve was linear in the range of 0.092μg-0.46μg/μ1 of Astragaloside.To study the pharmacokinetics of Astragalus, seventy mice ofKuriming species were equally randomized into two groups and each tookadministrated drug once a day for a week, each group was subdivided intosubgroups. The first three subgroups were group Ⅰ(with saline),groupⅡ(with artificial coradycepic hypha), group Ⅲ(with middle-dose of TotalSaponins of Astragalus).The second four subgroups were group Ⅳ(withlow dose of Total Saponins of Astragalus), group Ⅴ(with middle dose ofTotal Saponins of Astragalus), group Ⅵ(with high dose of Total Saponinsof Astragalus), group Ⅶ(with "Huangqijing" oralsolution).The resultsshowed that there were significant difference between the first threesubgroups, and between the four subgroups. The evidence show that even??low dose of Total Saponins of Astragalus can affect functions of T cells inmice.
