PART I The comparing analysis effect of anti-virus between rhIFNct23 ?b IFNaIb and IFNcIIb mutant and the induction MxA protein by IFNct2b Purpose : To study the anti-virus effect of various subtypes of the rhIFN from different manufactories and the induction of MxA protein in WI-38 cell by IFNGL2b. Methods: rnicrodose cytopathogenic inhibiton effect were applied to investigate the anti-virus effect.the WI-38 cell were induced by IFNcL2b for 3 6 12 24 hours.the contents of MxA protein in cytoplasma of induced W1-38 were detdcted by Western-blot. Results: In our experiments 5001U/ml, 501U/ml, 51U/mI of IFNa,a or IFNcL,b can prevent Wish cells from infecting by CQX B3 virus of 3 log4,21og4,llog4 TCID50 respectively ; Same doses of IFNs can also prevent Vero cells from infecting by both HSV-â… and HSV-â…¡ of 1000,100,10 TCID50 respectively; 5001U/ml,501U/ml, 51U/ml IFNcLIb or IFNaIb mutant can also prevent Vero cells from infecting by both HSV-I and HSV-II of 1000,100,10 TCID50 respectively; 1000 lU/mi IFNcc2b can cause expression of MxA protein in WI-38. Conclusion : Both the imported and domestic IFNs showed similar antivirus efficience on above 3 virus ;IFNα1b and IFNα2b mutant showed similar antivirus efficienee on above HSV-I and HSV-IL virus; MxA protein can be induced exclusively by IFNα2b; IFNα2b can cause expressin of MxA protein in WI-38.
