Oh/a ctlv. 1. To establish polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)combining silver stainning to detect Apolipoprotein E gene polymorphism. 2. To study the Apolipoprotein E(ApoE) genotypes and its alleles frequencies distribution of unrelated normal Chinese of Han nationality in Jiangsu area. 3. To detect the effect of ApoE polymorphism on serum levels of lipids,lipoproteins, apolipoproteins and its relationship with coronary heart disease(CHD).Methods: 1. 168 cases of normal Chinese of Han nationality in Jiangsu area and 63 cases of patients with coronary heart disease were collected. Exon 4 of Apolipoprotein E fragment coding for 112 and 158 amino acid residues was amplified by PCR, and the product was digested by Cfol endonuclease ,after electrophoresis and silver staining,the specific CfoI fragments of different sizes were visualized. 2 . The ApoE genotypes and its alleles frequencies distribution of unrelated normal Chinese of Han nationality in Jiangsu area were calculated,and were compared with other populations. 3. The effect of ApoE genotypes and alleles on serum lipids,lipoproteins,and apolipoproteins variation were analyzed. 4 . The genotypes of CHD(63 cases) were also determined, and their ApoE alleles frequencies were compared with control subjects(90 cases) from normal group whose age, sex were matched with CHD subjects.Results:1. The frequencies of the ApoE genotypes in normal Chinese of Han nationality in Jiangsu area were separatedly: ~ =0.60%; ~ = 11.90%; ~ 2/41.20%; E = 10.70%; c75.00%; E = 0.60%;and the frequencies of three3common alleles were separatedly: c 2=7.14%, ~ 386.31%, ?4=6.55%,there were no differences among the groups of various age and sex in the distribution of ApoE alleles frequencies. The distribution of ApoE alleles frequencies in Jiangsu area was similar to that of Chinese population in other area, but the E ~ allele frequency was lower than that in Europe and Amenca, while the ~ ~ allele frequency was higher.2. The effect of genotypes and alleles on TC,LDL-C, ApoB along a decreasing gradient was ?3/4> c 3/3> ~ 2/3 and c 4> e 3> e 2.The effect of e allele was to increase serum levels of TC,LDL-C and ApoB, and the ?2 allele had an effect opposite to that of E allele.3. The frequencies of E allele in patients with CHD(12.70%) was higher than that in control subjects(5.55%).Conclusioff 1. PCR-RFLP is a rapid and simple method for obtaining accurate and reliable result. 2.The frequencies of the three ApoE alleles of normal Chinese population in Jiangsu area is similar to those of Chinese population in other area, and the ApoE alleles are highly variable among different populations. 3.ApoE polymorphism is associated with serum levels of lipids and apolipoprotiens. The effect of c ~ allele was to increase serum levels of TC,LDL-C and ApoB, and the E 2 allele had an effect opposite to that of ?~ allele. 4.Apo e ~ allele is probably the genetic marker of CHD.
