One important reason of the periodontal ligament tissue damage is the abnormal function of the periodontal ligament flbroblast induced by abnormal occlusion pressure. To investigate the influence of pressure to the proliferation of human periodontal ligament fibroblast (HPLF), in this study we set up a unit of HPLF pressure model so we can load intermitten pressure on primarily cultured HPLF. The interaction of the function of HPLF and loaded pressure was studied and experimental result was as follows: Using MTT assay to determine the cell proliferation of primarily cultured HPLF under discontinuous pressure, the HPLF OD value of pressured groups is much smaller than the control group at different time . Various magnitudes and durations of the discontinuous pressure can significantly suppress HPLF proliferation with the magnitude-dependent. The Flow Cytometer was used to observe the influence of pressure to the cell circle, The pressure culture condition can restrain the proliferation and division of HPLF. Interleukin- 1 f3 is a kind of cytokine with multi biological activities. It stimulate osseo-absorption and play an important role in the damage of periodontal tissue. To observe the influence of occlusion pressure the production of IL-i f3 of HPLF, the ELISA method was used to detect the influence of pressure to primarily cultured HPLF. HPLF produce higher IL-i P under pressure culture condition. To investigate the influence of occlusion pressure on the cyclooxygenase-2 (COX-2) gene expression in human periodontal ligament fibroblast. Prepare COX- 2 cDNA probe by RT-PCR and label it with isotope , use dot blotting hybridization to observe the changes of COX-2mRNA expression. the COX-2 expression level of HPLF loaded intermittent simulated occlusion pressure is significantly higher than the control group with the magnitude-dependent.
