ObjectiveThe changes of HSP7(K TNF-a and oxygen free radicals appeared in body after trauma in normal environment. Should these factor be appeared special change combined with the hot and humid environment ? the aim of this study was to investigate the dynamic changes of HSP70> TNF-a and oxygen free radicals after being shot in gunshot wound on limbs of canine in hot and humid environment, and reveal the function of heat acclimatization. To provide the academic evidence of the treatment of gunshot wound in hot and humid environment.Materials and methodsEighteen mongrel canine were divided into three groups: trauma in normal environment (NE) , trauma in heat acclimatization (HA), trauma in hot and humid environment(HHE). There were 6 canines (6 hindlimbs) in each group. After being shot at the unilateral thighs where the muscle was full and round (avoiding the injury of bone and blood vessels) th^j canines were put into the controlled climate room. The study included 3 parts:1. The expression of HSP70 of lymphocytes was measured by Western dot blot in peripheral blood and muscle tissue in gunshot wound tract at different time point after wound.2. The expression of TNF-a were measured by ELISA in peripheral blood and muscle tissue in gunshot wound tract at different time point after wound.3. The expression of SOD and MDA were measured respectively by MTT and fluorescence in peripheral blood and muscle tissue in gunshot wound tract at different time point after wound.Results1. The changes of HSP70: There were two peaks in hot and humid environment after gunshot wound on limbs of canine, the first peak appeared at the time of 4 hour after wound . because of hot and humid environment, the second peak appeared at the time of 10 hour after wound. HSP70 at the time of 14 hour after gunshot in hot and humid environment group was significantly lower than that in other groups. Through the heat acclimatization, base level of HSP70 and HSP70 at the time of 14 hour after gunshot in hot and humid environment were significantly ascended.2. The changes of TNF-a: TNF-a was ascended in every group, The curve forms of the content of TNF-a were concordance. The maximum of TNF-a in every group was differential expression (p<0.05). TNF-a in HHA group was the highest, TNF-a in NE group was the lowest. These indicated that expression of TNF-a was increased by trauma or hot humid environment. Which combined.increased more. The TNF-a in muscle tissue in gunshot wound tract was significantly higher than that in the peripheral blood.3. The changes of Oxygen free radical:3.1. MDA: MDA in plasma ascended to the peak at the time of 10 h in NE group, ^he maximum and the level at 14-18 h were lower than that in other groups, P<0.05. But the extent of the ascended MDA was lower than that in HHA group. The level of MDA in muscle tissue in gunshot wound tract at the time of 18 h was concordance in three groups.3.2. SOD: SOD in erythrocyte ascended to the peak at the time of 8 h in HA group, and the level of SOD after the time of 6 h were significantly higher than that in HHA group, p<0.05. SOD in erythrocyte ascended to the peak at the time of 6 h in HA group. The level of SOD in erythrocyte after the time of 6 h was the lowest in three groups. SOD in muscle tissue in gunshot wound tract wasn't detection at the time of 10-24 h in HHAgroup, the time that showed similar pattern in other groups was at 14-24 h. the content of SOD in muscle tissue were lower than that in erythrocyte.Conclusions1. Both HHE and gunshot could induce the expression of HSP70, It played important roles and showed synergistic effects in stresses-tolerance.2. The TNF-a was positive correlation to the increasing of MDA in every group, that indicated TNF-a and lipid peroxidation showed synergistic effects in the tissue injuries.3. The SOD was positive correlation to the MDA in every group in period of 14 hours after gunshot wound in hot and humid environment, this result indicated stresses can induce expression of sup...
