Background and Purposes Oxygen radical has been implicated in the pathogenesis of atherosclerosis.NADPII oxidase,a major source of superoxide generation in neutrophuls and the vascular system,plays a critical role in ischemic injury and atherosclerosis.Recently, an association between the C242T polymorphism of p22phox,an essential component of NADPH oxidase,and cerebral infarction(CI) ,coronary artery disease and peripheral arterial occlusive disease in the world.But there were several conclusions about it. To investigate the C242T polymorphism of p22phox in Chinese,and the relationship between the C242T and cerebral infarction,we conducted a case-control study. Materials and Methods We recruited 129 Cl patients(67 atherothrombotic infardtion patients and 62 lacunar infarction patients)and 122 control subjects without cerebrovascular diseases or coronary diseases,who were admitted in Nan Fang I-Iospital.Genotypes were determined by polymerase chain reaction and restriction digestion with RSA I enzyme .Subjects with CC genotype(n205) were identified by the presence of 395bp and I I 3bp products on gel electrophoresis.Those with TT(n=45) were identified by the presence of 3 l6bp ,1 l3bp,and 79b products and those with CT(n1)by the presence of 395bp,316bp,l l3bp and 79bp products. Results The frequency of the C allele of the C242T gene was O.934,and the frequency of the T allele was O.066.The TC+TT genotype frequencies in the CI group and the control group were 24.0% and 13.3%,respectively,and the prevalence of the TC+TT genotype was significantly higher in the CI group( x 27.564,p 0.OO3, 0R2.304, 95%CI l.25-4.23).Analysis by CI subtypes showed that the frequence of CT+TT genotype was 28.4% in the atherothrombotic infarction patients,while 19.4 % in the lacunar infarction.There was a significant difference between the atherothrombotic patients and control group( x 2=7.564,p0.003, 0R2.304, 95%CI 1.25- 4.23) ;but no significant difference between the lacunar infarction patients and the control group,although the frequency of TC+TT was higher than the control group.When we compared the frequency of the CT+TT genotype of the atherothrombotic group with the lacunar infarction group,there was no significant difference ( x 2 l 430 p=O 201, 0R 1.57 95 %CI, 0.79-3.15) The TC+TT genotype frequencies in hypertension patients and the patients without hypertension were 47.4% and 15.6%,respectively,and the prevalence of the TC+TT genotype was significantly higher in the hypertension patients( X 2 16.72,
