Hypertrophic scar is one of the most common pathological phenomenon seen after wound healing of trauma, especially in deep burned patients, or say it overhealing of wound .It raises above skin level significantly, in most case with the contracture of scar, which causes serious deformity and dysfunction, and worse the effects of rehabilitate and quality of life for patients. The mechanism of its formation is still in debating, by now we have reach consensus on it .the fibroblasts play an important role in the process.Up to now, there is no one satisfied method that can deal it all right, prevention is still the priority. Several treatment modalities exist. They are: Surgery, Pressure ,Silicone Materials, Radiation , Cryotherapy, Corticosteroids, Laser and other ways .combined ways can get better results than the sole one .But they all have some trauma to patients , or with big side effects ,and can not be used widely with some limits ,and what's more ,they can hardly provide satisfied effects . We observed that the hypertrophic scars usually undergo at least partial spontaneous resolution over widely varying time courses. So we save no efforts in finding the very drug that accelerates the process to curb it.lt has been proved that the main character of hypertrophic scar is excess matrix production. Exocytosis is a crucial calcium ion-dependent step in the secretion of extracellular matrix macromolecules.If secretion is blocked, biosynthesis is inhibited. In the past few years, we found that calcium ion in the hypertrophic scars is higher, and calciumantagonists such as verapamil can inhibit the proliferation of fibroblasts derived from it and synthesis of extracellular matrix, and ease contracture extent, and has gotten better results in clinical trial..Tetrandrine is one extrat of hanfangchin, we find that it has effect acting like calcium antagonist, and has gotten satisfied effects on the treatment of silicosis . it is one of basic drugs in the treatment of silicosis in our country. By now. it is testified that it slao has good results on the treatment and prevention of hepatic fibrosis and glomerulus fibrosis, mostly by inhi -bitting proliferation and decreasing sythesis of extracellular matrix. Seen as one of fibrosis diseases on some extent, with over proliferation of fibroblasts as character, we study its effects on fibroblasts derived from hypertrophic scars in vitro to explore its value on the treatment of hypertrophic scar.In the experiment, we have fibroblasts cultured in five different concentration of Tet 0,10,20,40,80ug/ml,and undergo MTT, 3H-Tdr, test cell circles and the concentration of LDH in the supernatant to find its cellular toxicity. We find that Tet can inhibit the proliferation of fibroblasts derived from hypertrophic scars ,and the highest inhibitive ratio is 72.1% when the concentration reaches 80ug/ml, and decrease DNA synthesis and the proportion of S stage in cell circle, under the concentration of 80ug/ml, it doesn't alter cell shape. Statistically ,there is no significant difference of the LDH concentration in the supernatant between these groups. So we assert it has no significant cellular toxicity under 80mg/L. It accomplishs this role mainly through inhibitting DNA sythesis. From this experiment, we draw the conclusion that Tet can inhibit proliferation of fibroblasts derived from hypertrophic scars in vitro, it may has some effects on prevention andtreatment of hypertrophic scar.
