| Objectives To investigate the adhesion of endothelial cells (EC) to polymorphonuclear leucocytes (PMN) from patients at different phase of cerebral infarction (CI) and the adhesion of PMN to hypoxia/ reoxygenation(H/R)-stimulated endothelial cells. Furthemore, to define the effects of antibodies to intercellular adhesion molecular-l(ICAM-l monolonal antibodies, anti-CD54 antibodies) upon the adhesion. Methods We compared PMN interaction with human umbilical vein endothelial celIs-304(HUVEC-304) with and without anti-CD54 antibodies. Then we compared PMN interaction with H/R-stimulated endothelial cells with and without anti-CD54 antibodies. Results 1 , Adhesion of PMN from normal people to VEC-304 was reduced by Anti-CD54 antibodies (P<0. 05) . 2, Adhesion of PMN from patients of CI within 1 week to VEC-304 increased evidently (P <0.01) , but adhesion of PMN from patients with CI at third week to VEC-304 became normal ( P , 0.05 ) . 3 , Anti-CD54 antibodies reduced adhesion of PMN from patients with CI within 1 week to VEC-304 increased highly ( P < 0.01 ) , but anti-CD54 antibodies reduced adhesion of PMN from patients with CI at third week to VEC-304 slightly ( P < 0.05 ) . 4 , Compared with NonH/R-stimulated endothelial cells , adhesion of PMN from patients with CI to H/R-stimulated VEC-304 increased highly (P<0.01) , and adhesion of PMN to H/R -stimulated VEC-304 was 4 times than adhesion of PMN from normal people. 5 , Adhesion of PMN from patients with CI within 1 week to H/R-stimulated VEC-304 was reduced to 45.20±3.04% from 79.4 + 2.93% by anti-CD54 antibodies, and adhesion of PMN from patients with CI within 1 week to Non-H/R-stimulated VEC-304 was reduced to 27.80 +2.64% from 41.53 +3.09% by anti-CD54 antibodies. 6. Hyperlipid did not activated PMN from patients with CI within 1 week, but hypertention and diabetes enhanced activation of PMN.Conclusion 1 . PMN is involved in cerebral injury after cerebral infarction. 2. Adhesion of PMN to endothelial cells can be mediated by ICAM-1 (CD54), anti-CD54 antibodies can partially block the adhesion. 3 , Hypertention and diabetes have effects on activating PMN . |
