Impact Of Chinese Herb Qingyuan Capsule On LDLR Gene Expression In Rats Suffering From Hyperlipidemia

Objective To study the relationship between hyperlipidemia and low-density lipo-protein receptor (LDLR) gene mRNA expression in hepatic cells of rats, as well as the effect of Qingyuan Capsule on LDLR gene.Method Wistar rats were selected and divided into 3 groups: Control, hyperlipidemia group and treated group with Qingyuan Capsule . The low-density lipoprotein receptor (LDLR) gene mRNA expression was detected in all of Wistar rats with semi-quantified RT-PCR , respectively.Result In normal hepatic tissue, 266 bp DNA band is visible, which indicates that LDLR was expressed normally. However, in hyperlipidemia group, this kind of hepatic tissue expressed less amount of LDLR compared to normal tissue. Furthermore, through analysis of statistics, the difference was significant (P<0. 05). After the administration of Qingyuan Capsule for a period in hyperlipidemic rats, expression of LDLR gene almost restored up to the similar level with that of normal group, while the untreated group has no significant change in terms of LDLR gene expression.Conclusion The expression of LDLR in hyperlipidemic rats, compared to normal rats, is dramatically decreased. Qingyuan Capsule has the capacity to induce significantly the LDLR expression.
