Objective To investigate the expression of PTEN and cyclin E in human gastric carcinoma in relation to biological behavior and clinical significance. Methods The expression of PTEN and cyclin E in 106 cases of resected human gastric carcinoma was analyzed by immunohistochemical S-P method. The relationship among PTEN or cyclin E expression, various clinicopathological factors and the prognosis of gastric carcinoma was analysed. The relationship between the expression of PTEN and cyclin E was investigated as well. Results 1 <> The expression of PTEN in gastric carcinoma and its clinical significance.(1) Normal gastric mucosa was positive for PTEN, but a relatively low rate was found in carcinoma .Of the 106 patients with gastric carcinoma, 72(67.9%) showed positive staining of PTEN in the primary lesion. PTEN expression was low in 15(14.1%) and high in 57(53.8%) , respectively .(2) In gastric carcinoma, expression of PTEN inversely correlated with depth of invasion , pTNM stage and positively correlated with histological grade (PO.05), but its relation to sex , age ,lymph-node metastasis and remote metastasis was not significant.(3) The 3-and 5-year overall survival rate in the positive PTEN expression group was 48.6% ,37.5% respectively and 32.4% , 11.8% in the negative group (P>0.05). These data indicated that PTEN was not clearly associated with prognosis of gastric carcinoma.2^ The expression of cyclin E in gastric carcinoma and its clinical significance.(1) Normal gastric mucosa was almost negative for cyclin E ,but a relatively high rate was found in carcinoma.Of the 106 patients with gastric carcinoma, 52 (49.1%) were expressed positively for cyclin E. Cyclin E expression was low in 30(28.3%) and high in 22(20.8%), respectively.(2) In gastric carcinoma, expression of cyclin E inversely correlatedwith histological grade and positively correlated with pTNM stage(P<0.05), but no correlation to sex ,age , depth of invasion ,lymph-node metastasis and remote metastasis was significant.(3) The 3- and 5- year overall survival rate in the positive cyclinE expression group was 25% , 15.4% and in the negatine group was 55.6%, 42.6%, respectively . There was significant difference in these two groups(P=0.0243). These data indicated that cyclin E was inversely associated with prognosis of gastric carcinoma.3 PTEN expression level inversely correlated with cyclin E expression level in gastric carcinoma (P<0.01).4x On multivariate analysis by Cox regression model , neither PTEN nor cyclin E expression level could be found to be of independent prognosis significance.Conclusion These data suggest the PTEN expression correlate with cyclin E expression and both may contribute to the genenis and progression of gastric carcinoma .Cyclin E may be a useful prognostic indicator in patients with gastric carcinoma.
