| Objective In order to investigate the situation of EBV infection in nasopharyngeal lymphoma and in latent state of EBV produce the viral oncogene protein LMP-1 which whether or not has the relationship with this kind of lymphoma. The same as others malignant tumor, lymphoma break the balance between the process of cellular proliferation and apoptosis, so make cell malignant growth and become immortal and then forming tumor. So we generally research this two different aspect to primarily find out the nasopharyngeal lymphomagenesis and progress course.Methods In this study, we use the EBER1/2 mRNA probe to test the EBV infection in 70 cases of nasopharyngeal lymphoma and 10 cases of nasopharyngeal chronic inflammation tissue with ISH method. Also we use IHC staining technique to mark the positive cells with CD20^ CD45RCK CD56^ LMP-K PCNA^ CyclinDK Bcl-2, FaSx FasL, and analyse the correlation between each other to make certain function in forming nasopharyngeal lymphoma. Simultaneously we apply the TUNEL technique to quantitatively take count of the apoptotic cell.Result The positive rate of the infection of EBV in nasopharyngeal lymphoma (62.9%) is higher than that in nasopharyngeal chronic inflammation tissue (0%) (PO.0001). Similarly protein LMP-1 also has different expression between this two different pathological changes (68.6%., 20%), tumor tissue has the remarkable higher expression. In this research there's 12 cases NK/T cell lymphoma, shows positiveexpression of CDS 6, which have the relation with protein LMP-1, but have no specital biological behavior. Furthermore some proliferation and apoptosis indexes were investigated, such as: Cyclin Dl> PL Bcl-2> Fas^ FasL and AI. In both rumor and inflammation tissue, there were obvious difference, hi addition, the positive expression of protein LMP-1 has correlation with appearance of clinical B symptom, and with no relationship with clinical stages ^ region lymph node intumescence, but show significance in prognosis of nasopharyngeal lymphoma. Also all the indexes of proliferation and apoptosis have no connection with LMP-Tpositive expression. Moreover, in the analysis of the correlation of these indexes, show that the expression of Cyclin Dl is in connection with PI (P=0.001). Other investigations were taken and indicated in B cell lymphoma has the protein Bcl-2 higher expression and in NK/L cell lymphoma has protein Cyclin Dl higher expression. Survival analysis show that patient's age ^ region lymph node intumescence ^ treat manner first period of treatment and positive level of the expression of Fas protein-theses factors are very important for indicating the prognosis.Conclusion During the course of nasopharyngeal lymphomagenesis, EBV infection and protein LMP-1 expression make truly important action, and the expression of LMP-1 correlate with aggressive clinical behavior and prognosis, as well as CD56+ NK/T cell lymphoma. Our results show the abnormal expression of all proliferation and apoptosis indexes in nasopharyngeal lymphoma tissue, thereby affirming their functions. Because these indexes with no relationship with the expression of protein LMP-1, so we think in the process of lymphomagenesis, there is a complex starting signal transmit system, in which LMP-1 protein may activate different approaches to induce relevant protein expression, so indirectly make effects to form tumor. |
