Objective To explore new agents for treating pulmonary fibrosis ,the effects of marine Sulfated Polysaccharides 910 on Human Lung Fibroblast (HLF proliferation, basic fibroblast growth factor(bFGF)-induced HLF proliferation and DNA/ protein synthesis were investigated in this paper.Methods The effect of SP910 on proliferation of cultured HLF was evaluated by MTT assays. And the result was compared with control group. Furthermore, the inhibitory effects of SP910 on bFGF-mediated HLF proliferation was observed. We also investigated the effect of SP910 on DNA and protein synthesis of HLF.Results A dose-dependent anti-proliferative effect of SP910 was observed. The results indicated that SP910 inhibited HLF proliferation at concentrations from 5ug/ml to 160ug/ml, with its maximal inhibitory rate (84.65%, P<0.001) at concentration of 160ug/ml. Moreover, the results show an inhibitory effect of SP910 on bFGF-mediated HLF proliferation in a dose-dependent manner, but the anti-proliferative effect of SP910 on bFGF-induced HLF proliferation decrease with the increasing of bFGF doses. SP910 showed no anti-proliferative effect at bFGF concentration of 10ng/ml. The results determined by Flowcytometry indicated that SP910 may inhibit DNA and protein synthesis of HLF.Conclusions This study suggest that SP910 have anti-proliferative effect on HLF and this effect may result from blocking bFGF-HSPGs-bFGFR complex formation, which is important for bFGF signaling.
