Objective:To investigation the association of the Apo E polymorphism and gallstone in type 2 diabetes mellitus .Methods: We selected 33 type 2 diabetics with gallstones(GSD+) ,38 type 2 diabetics without gallstone(N-GSD) and 60 normal controls. Rapid-cycle PCR and fluorescence resonance energy transfer (FRET)with the Lightcycler was used to determine their Apo E genetypes.Results: (1 )FBG,Fasting plasma C-peptide, Tc and LDL-C in GSD+ were higher than those in N-GSD(P<0.01-0.05),while HDL-C in GSD+ was lower than that in N-GSD(P<0.01 ).(2)GSD+ had a lower frequency of Apo 3/3 genetype and 3 allele than those in control(P<0.05),but had a higher frequency of Apo 4/3 genetype and 4 allele than N-GSD(P<0.05)and control(P<0.05).(3)The incidence of gallstone in Group 4/3 3/3 was significantly higher than that in Group 2/2 3/3(P<0.01) and Group 3/3(P<0.05).LDL-C in Group 4/3 3/3 was significantly higher than that in Group 2/2 E 3/2 and Group 3/3(P<0.01), and Tc was also significantly higher than that in group 3/3(P<0.01).(4)Logistic regression showed that BMI,FBG,HDL-C and Fasting plasma C-peptide were the risk factors for diabetics with gallstone.Conclusions:The disorder of Lipoprotein metabolism,BMI,FBG and Fasting plasma C-peptide are risk factors for diabetics complicated by gallstones.The ApoE polymorphism was associated with the disorder oflipoprotein metabolism,and the presence of Apo 4/3, 4/4 genetype may be the genetic marker of diabetics complicated by gallstone.
