Tumor's occurrence and tolerance for treatment is highly concerned with the inhibition of apoptosis (APO). As compared with the necrosis, the APO was needed less stimulation, has a good integrality of cell membrane, and the formed apoptotic body was immediately phagocytosed by the ambient phagocytes and has less affection to the organism. Therefore inducing APO has become to the new mode of treating tumor. To treat the tumors with the traditional Chinese medicine is a peculiarity in domain of tumor treatment in China. Many clinical practices had proved that the traditional Chinese medicine alone or combination of Chinese traditional and Western medicine has certain curative effects for the treatment of tumor and could also relieve the side effects of radiotherapy and chemotherapy. It was reported that valid component of compound or sole the traditional Chinese medicine could induce the apoptosis of tumor cells. The Rubia Cordifolia L is one of the traditional Chinese medicines. It was recently reported in Japan that effective component of the extract of Rubia Cordifolia L has obvious inhibition for leukemia, cancer of colon and carcinoma of the lung in mice, its curative effects equaled or exceeded to the catharanthine, mitomycin and adriacin and had less virulence for normal cells. But there were no reports about its effects for the gastric carcinoma.Using the MTT assay, agarose gel electrophoresis, transmission electron microscope, TUNEL method and immunohistochemistry (PCNA, bcl-2),it was studied in this experiment that the inducing APO, anti-proliferation and affection of expression of bcl-2 gene before and afterAPO of the Rubia Cordifolia L with the 5-fluorouracil(5-fu) for MGC-803 cells of gastric carcinoma. The experiment results showed as follows: (1) 0.2~1.0mg/ml of the extract of Rubia Cordifolia L and 2~16(g/ml of 5-fu had inhibition for MGC-803 cells of gastric carcinoma and there had relationship of concentration dependence, the inhibition rate between the different concentrations had significantly different (P <0.05 or P <0.01) and was obviously increased when using two combined medicine (P <0.05 or P <0.01or P <0.001) ; (2) The microvillus of APO cells disappeared, cytoplasm had vacuolation, and chromatin was pyknosis and coagulated to mass clustered around the nuclear membrane by transmission electron microscope; (3) The normal control group only had a large molecular band very near by adding sample foramen by agarose gel electrophoresis of DNA, but the medicine administrated group had incontinuous trapezoidal band; (4) 0.2, 0.4, 0.8 and 1.0mg/ml of the extract of Rubia Cordifolia L and 2, 4, 8 and 16(g/ml of 5-fu respectively induced APO, inhibited the cell proliferation and had the relationship of concentration and time dependence, and there had significant difference between different concentration and affective time (P <0.05 or P <0.01) except there was no different between the individual concentration and time(P >0.05) . The % of APO of MGC-803 cells was increased and the positive expression rate of PCNA was decreased (P <0.05 or P <0.01) when using two combined medicine; (5) After administrating the 0.8mg/ml of the extract of Rubia Cordifolia L to MGC-803 cells for 48h, the positive expression rate of bcl-2 was obviously decreased (P<0.001).The experiment results indicate that (1) The extract of Rubia Cordifolia L has the effects of anti-proliferation and inducing APO; (2) Whenadministrating with 5-fu, the extract of Rubia Cordifolia L obviously enhanced the effects of anti-proliferation and inducing APO of 5-fu; (3) The basement of molecular biology of the extract of Rubia Cordifolia L for inducing APO of MGC-803 cells is correlated with inhibiting the expression of bcl-2 of anti-apoptosis gene.
