As a new type information technology, biosensor exerted important role in the field of biomedicine. The fluorescent & fiber optical biosensor for determination of cholesterol is a important medical biosensor which has comprehensive prospect of application.As a excellent material for preparation of biomembrane, cellulose acetate (CA) is a fine carrier for the immobilization of cholesterol oxidase (COD) . In this dissertation, cellulose acetate was selected as immobilization carrier for COD and the CA membrane was prepared by pump means and salt leaching, the catalytic activity of immobilized COD based in the membrane and some external factors, such as characteristic of membrane, immobilization condition and reaction condition were detailed studied. In order to discuss the application of sol-gel in the preparation of biomembrane, the gel membrane of agarose was prepared by sol-gel with acticarbon and agarose as raw material, at the same time, the catalytic activity of immobilized COD based in the membrane was studied.A high effect fluorescent reagent which is Ru(phen)32+ was prepared with hydration ruthenium trichloride and 1,10-phenanthroline as raw material, at the same time, the crystal structure , violet & visible spectrum and fluorescence spectrum of this reagent was studied. A fluorescent reagent membrane was prepared with Ru(phen)32+ as raw material and cellulose acetate as carrier, after the study of oxygen's quenching function for this fluorescent reagent, the extreme sensitive oxygen quenching function of this fluorescent reagent and its characteristic of fitting the detection of hypoxia environment were discovered.After study on the technology of probe head of the optical fiber, a biosensor for determination of cholesterol which based on fluorescence quenching and adopted phase shift & phase lock technique to detect the change of light intensity was developed, in this biosensor, the cellulose acetate COD enzyme membrane was took as sensitivity basic dollar, Ru(phen)32+ was took as indicator of oxygen and furcated optical fiber as conduct carrier of light signal. In this dissertation, the characteristic of this sensor in determination for gas oxygen and dissolved oxygen was studied, thereafter, the testing of cholesterol solution and plasma also was developed in experiment and obtained ideal result.This sensor has higher sensitivity and excellent stability, in the detection of gas oxygen and dissolved oxygen, the response time were 5.57s and 14.625s.
