Object It is useful to treat a variety of neuralgic disease by neuronal transplantation. The recent discovery demonstrated MSCs will be ideal cell resource to neuronal transplantation. To investigate whether MSCs could be induced to differentiate into neuron-like cell by ATRA, MSCs collected from adult humen and rats were treated by ATRA or ATRA with different growth factors and chemical reagents. These experient here will offer importment basement to apply MSCs to neuronal transplantation.Methods MSCs were treated by the 7 following groups of neuronal differentiation conditions: group 1: 1 mmol/L BME ; grpup 2: 0.5 μmol/L ATRA; group 3: 10 ng/ml bFGF ; group 4: 0.5 u mol/ L ATRA+10 ng/ml bFGF, all above 4 goups were treated for 1 day; group 5: 0.5 μ- mol/ L ATRA for 2 days, added 5mmol/L BME to 5 groups respectively; group 6: 0.5 μ mol/ L ATRA for 1 day, then added 200 μmol/L BHA+2% DMSO to every group; grpup 7: 0.5 μmol/ L ATRA for 1 day, then withdrew ATRA, to observe change of morphology and number of cell during differentiation. Immunocytochemistry was used to detect the expression of NSE, NF-M, GFAP.Result MSCs could be induced to differentiate into the cells with a typical neuronal morphology under the every group of conditions, expressing intensive NF-M and increased levels of NSE, under 4th condition,5almost 80% of the cells expressed for NF-M,86% cell forNSE.Conclusion ATRA or ATRA with different growth factors and chemical reagents can induce MSCs to differentiate into neuron-like cell .There are different percentage of induced differentiation under different conditions of neuronal differentiation. MSCs were synergically treated by ATRA and bFGF, can get higher percentage of induced differentiation than that of other reports.
