Objective: To investigate the influence of XQM on chronic bronchitis mouse's T lymphocyte subpopulation in peripheral-blood, the weight of spleen ,the indexes of spleen and the dendritic cells ,and investigate the effective mechanism of XQM, and provide data for further study and clinical practice.Methods: 66 healthy ICR mice including 33 male and 33 female were divided into two groups in random. Ten mice were the normal control group(Group A).The experimental group ,50 mice were administered ammonia to induce chronic bronchitis. Then they were divided into 5 groups in random according to their weights and each group had 10 mice including 5 male and 5 female. B was chronic bronchitis model group. C was higher dose XQM-treated group, D was lower dose XQM-treated group, E was positive control group, which was administered nucleic acid and casein hydrolysate composite oral medicine (NACH), F is XQM&NACH-treated group.After experiment, by observing the pathological change of bronchitis, by detecting the weight of spleen and indexes of spleen, by using immunohistochemical method to detect the change of CD3+, CD4+,CD8+ and the activity of S-100+ cells in the mice's peripheral blood, and by statistical treatment and comparison, investigate the curative mechanism of XQM.Results:(1)In the observation of pathologic appearance, there is less inflammatory harm in XQC-treated group than the chronic bronchitis model group.(2)Compared with the chronic bronchitis model group, the weight and indexes of spleen are increased in two XQM-treated groups and XQM&NACH-treated group.(3) Compared with the chronic bronchitis model group, the total T lymphocytes (CD3+) are increased significantly (p<0.01),and the level of CD4+, CD4+/CD8+ is increased significantly in two XQM-treated groups, and XQM&NACH-treated group (p<0.01), and there is the obvious relationship between the dose and the effect.(4) Compared with the chronic bronchitis model group, the activity of S-100+ cell is decreased significantly in two XQM-treated groups, NACH-treated group and XQM&NACH-treated groupConclusions:(1) The observation of the pathologic appearance shows: XQM has the obviously therapeutic efficacy to the inflammatory harm in chronic bronchitis mice, can alleviate the inflammatory reaction in chronic bronchitis mice.(2) Xianqi mixture can strengthen the function of immunity system and increase the weight and indexes of the immune organ : spleen. The total T lymphocytes (CD3+) are increased significantly in XQM-treated groups (p<0.01). There is obvious relationship between the dose and the effect.(3)After taking Xianqi mixture the dendritic cells in mice's lung tissue are decreased significantly compared with the chronic bronchitis model group(p<0.01). The result shows that Xianqi mixture has anti-infection function. |
