Therapeutic drug monitoring(TDM) have been experienced for more than 20 years in the our country. Many scholars have determinated blood or urine drug concentration with some kinds of theories and techniques in this period. Since the 80's , the study on the pharmacokinetics of Chinese herbal medicine and compound has been continously reported. In the field, "Pharmacokinetics of traditional Chinese medicine syndrome and recipe (RS-PK)"put forward by professor Huang Xi,includes traditional Chinese syndrome PK(S-PK) and traditional Chinese recipe PK(R-PK). In the strategy, Prof. Huang brought forward the idea of traditional Chinese medicine recipe therapeutic drug monitoring (R-TDM). Traditional Chinese medicine(TCM) has not formed their characteristic PK as a consequence of unknowning that what are chemical components absorbed into the circulation after administeringtraditional Chinese recipe(TCR). Then, our progresses in successful detection of TCR-derived component in blood supported the above-mentioned idea. The more and more determinations of in vivo TCR-derived compound laid the direct foundation of advancing the idea.we choose Suxiao Jiuxin Wan(SXJX) and its chemical components ?bomeol and isoborneol as the examples to test the above -mentioned idea. The same dose of drug was used in the study on urine drug concentration of blood stasis syndrome -- traditional Chinese syndrome(TCS) and healthy groups. To set forth the classification, terminology, concept and significance of the hypothesis and provide the evidence for it in the present study.Methods:The following main instruments were used in the experiment:Gas chromatography instrument HP4890D with HP-5 quartz spring capillary column.Chromatographic condition:Determination of urine borneol and isoborneol concentration桹VEN TEMP 75'C, INJA TEMPI40癈, DETB TEMPI 50癈, The velocity of flow is a 8.1ml/min.Determination of blood bomeol and isoborneol concentration 桹VEN TEMP85'C, INJATEMP230癈, DETB TEMP 240癈; The carried gas was nitrogen gas year (N2) , The velocity of flow is a 8.3ml/min , Hydrogen ion flame detector (FID) .Samples:The blood /urine samples of six coronary artery disease with blood stasissyndrome patients and six healthy volunteers were obtained from decapitation after a single oral SXJX (10mg/kg)at 10,30,60and 90min, respectively.Sample preparation: Blood sample contained bomeol and isoborneol was solved with skellysolve B:trichloromethane (10:l)solution. Urine sample contained bomeol and isoborneol was solved with skellysolve B:Ethoxyethane(l:l)solution. internal standard of Naphthalene. And then 3 # 1 was determined by GC each time.Results:The results of borneol urine concentration after SXJX at 10,30,60 and 90min are as follows:In healthy volunteers 1.148 ?0.240,2.766 ?1.731,5.982 ?4.178,5.905 ?4.614 # g/ml;In blood stasis syndrome patients 1.056 ?0.549,4.939 ?2.165,8.174 ?5.327,5.686 ?3.509 u g/ml.Through this experiment we found the significant differences in borneol urine concentrations between healthy volunteers and blood stasis syndrome patients(p<0.05).The results of borneol plasma concentration after Oral SXJX at 10,30,60 and 90min are as followsrln healthy volunteers 15.82 ?9.890,18.68 ?14.55,10.63 ?4.49,6.16 ?1.82# g/ml;In blood stasis syndrome patients 55.65 ?17.89,41.07 ?32.69, 28.17 ?11.54,12.83 ?6.33 # g/ml.Through this experiment we found the significant differences in borneol plasma concentrations between healthy volunteers and blood stasis syndrome patients(p<0.05 ).The state of the syndrome in blood stasis syndrome affected obviously the absorption, distribution, metablism and excretion of borneol and10isoborneol.Conclusions:1 This experiment first study the method of determinate Borneol in urine man after oral SXJX.The methods for the determinations on borneol and isoborneol urine concentrations were simple, rapid, sensitive, accurate, especially and reduplicative.
