The effect of anti-allergic asthma of SCZ(SFE extract of Fructus Cnidii)and its mechanisms were investigated in this paper.The latent period of asthmatic model in guinea-pig induced by inhaling 0.15% His or 0.1% His and 2% Ach were prolonged after administration of SCZ.In the isolated guinea-pig tracheal experiments, the direct relaxant effect.the inhibitory effects of SCZ on 2 10-5M His,2 10-5M Ach or 3 10-5 KCl-induced contraction were observed .The dose-effect curve of His was depressed significantly by SCZ. The increase of time of perfusate escaping from the lung surface in mice induced by Ach was inhibited by SCZ.The allergic asthma reaction in guinea-pig challenged by inhaling ovalbumin was also suppressed by SCZ. The significant reduction of NO of BALF or lung, content of BALF protein, MDA and MMS in serum were found in sensitized asthmatic model in rats after administration of SCZ.It was also observed that the PCA in mice or rats, the ear edema induced by bimethylbenzene were inhibited by SCZ. The relaxant effect of SCZ on isolated ileum strip in sensitized guinea-pig induced by ovalbumin precontracted with ovalbumin was also found.AbstractThe contraction of isolated guinea-pig ileum induced by His, Ach , 5-HT was antagonised by SCZ (IC60=3. 72 10-2mg/ml, 6. 46 10-5mg/ml, 4. 26 10-2mg/ml, respectively) . The histamine or 5-HT-induced cutaneous capillary permeability was suppressed . The licking response induced by 4-AP in mice , the mast cell degranulation of skull in rats could be inhibited significantly. Moreover, The degranulation of peritoneal mastocyte in rats also be suppressed by the serum containing SCZ.It was also found that the MDA generation and malondialdehyde generation from phospholipid in vitro could be inhibited significantly by SCZ.The delayed hypersensivity reaction resulted from DNCB was suppressed significantly. In addition, the spleen index and the thymus index wasn' t changed by SCZ at dose of 400mg/kg, 200m g/kg. The expectorant effects was observed in mice.These results indicated the effect of anti-allergic asthma of SCZ may be related to tracheal-relaxantion, antagonism of allergy mediators, immuno-modulation, anti-oxygenation, anti-inflammation, expectorantion effects.
