ObjectivesAlzheimer's disease is a progressively degenerative disease in the central nervous system, and its etiological factors and pathogenesis are not very clear. In our study, we used compound AD model rats by damaging both sides of NBM with IA after injecting D-galactose into the abdominal cavity to observe the influence of Tiaoxin prescription and Zishen prescription on the learning and memory abilities and activities of CHE and MAO. We discussed the mechanism of the treatment for AD with Tiaoxin prescription and Zishen prescription.Methods1 Preparation of subacute aging model rats: 1% D-galactose was injected into the abdominal cavity of Wistar rats, 50mg/kg/d for 6 weeks.2 Preparation of AD model rats: The anesthetized rats were fixed on the brain stereotaxic apparatus. Then we used IA to damage both sides of NBM according to the stereotaxic atlas of rats.3 Learning and memory test by Y maze: We used the trisected Y maze to test the learning and memory abilities of the rats and recorded the number of the times of electric stimuli until 9 correct responses in 10.4 Determination of the activities of CHE and MAO: We used spectrophotography techniques to determine the activities of CHE and MAO in the hippocampuses of rats.5 Statistical analyses: SPSS 11.0 was used to analyze all the data. Thelearning and memory abilities of different groups were compared by pant-test before and after the treatment of different drugs. The activities of CHE and MAO were analyzed by one-way analysis of variance, a = 0.05, two-sided.Results1 Before and after the preparation of subacute aging models and comP<0und AD models, the learning and memory abilities of the rats decreased significantly (P<0.01, P<0.01) ; the activity of CHE decreased significantly(P<0.01, P<0.01), but the activity of MAO increased significantly (P<0.01, P<0.01) .2 After the subacute aging model rats were treated with Zishen prescription, Tiaoxin prescription and pracetam, the learning and memory abilities increased, and the memory ability increased significantly (P<0.01, P<0.05, P<0.05) ; the activity of MAO decreased significantly (P<0.01, P<0.05, P<0.05) , and the effect of Zishen prescription was more significant than those of Tiaoxin prescription and pracetam (P<0.01, P<0.01); the activity of CHE increased significantly (P<0.01, P<0.01, P<0.05) .3 After the comP<0und AD model rats were treated with Zishen prescription, Tiaoxin prescription and pracetam, the learning and memory abilities increased. The effects of Zishen prescription and Tiaoxin prescription on the learning ability were significant (P<0.01, P<0.05) , and the effects of Zishen prescription, Tiaoxin prescription and pracetam on the memory ability were significant (P<0.01, P<0.01, P<0.05) ; the activity of MAO decreased significantly after the treatment with Zishen prescription and Tiaoin prescription (P<0.01, P<0.01) , and the effect of Zishen prescription wasmore significant than that of Tiaoxin prescription (P<0.01); the activity of CHE increased significantly (P<0.01, P<0.01, P<0.01) , and the effects of Zishen prescription and Tiaoxin prescription were more significant than that ofpracetam (P<0.05, P<0.01) .Conclusions1 The comP<0und AD model by damaging NBM with IA after injecting D-galactose was close to the complicated clinical pathology of AD.2 After the subacute aging model rats and comP<0und AD model rats were treated with Zishen prescription, Tiaoxin prescription and pracetam, the learning and memory abilities increased, and the memory ability increased significantly; the MAO activity decreased, but the CHE activity increased; the effects of Zishen prescription and Tiaoxin prescription were more significant than that of pracetam, and the effect of Zishen prescription was most significant.3 The mechanism of Zishen prescription and Tiaoxin prescription may be relating to the effective improvement of the cholinergic system and adrenergic system of the brain.
