Objective To observe all-trans retinoic acid ( ATRA ) having effect of inhibiting proliferation of rat vascular smooth muscle cells ( VSMC) or not frome investigating the effect of ATRA acting on proliferation of VSMC and the expression of CyclinE,P21.Method Vascular smooth muscle cells were cultured by collagenase (primary culture) and trypsin (passage). 1×105 VSMCs were inoculated in 24 hole board and cultured with 5% CO2 in 37.DMEM without FBS will be used to enable cells go into GO Three groups were divided:(1)contrast group (n=5) ,cultured without any interfering.(2)FBS group (n=5) , cultured with 20% FBS. (3)ATRA group (n=5) cultured with 20% FBS and 2.5×10-6mol/L ATRA.After 24h,DNA synthesis 3H-TdR and CyclinE,P21 Western-Blotting were determined in order to observe the effect of ATRA acting on DNA synthesis and expression of CyclinE,P21 .All experiment date was express with x±s.Analysis of variance and t-test will be used between Difference groupes.Result (1)DNA synthesis was clearly inhibited in ATRA group compare withFBS group seen from 3H-TdR ( 3725.48±643.77 vs 11567.09±1974.67,P<0.01) . (2)The expression of CyclinE was clearly inhibited by ATRA seen from gel gray(122.48±3 53 vs 142.93±10.57,P<0.05 ) .(3)The expression of P21 was clearlyenhanced by ATRA seen from gel gray ( 151.40±9.98 vs 56 33±11 .06,P<0.001 ) .Conclusion (1) ATRA has effect of inhibiting proliferation of VSMC by reducing DNA synthesis and inhibiting the expression of CyclinE and enhancing the expression of P21. (2) It deduce that ATRA has effect of eliminating As,preventing stricture again after PTCA and stent surgery. (3) It prompt that ATRA' s effect of inhibiting proliferation of VSMC may relevant to inhibit the expression of CyclinE and enhance the expression of P21.
