Part one: Methods :We filtrated the learning ability decreaed rats from aged rats by three arms maze. 5-BrdU was used to label the proliferation cells. TUNEL was used to label the apoptotic cell .Then we count the positive cells labeled for 5-BrdU or TUNEL in the granule cell layer and hilus. Results:Comparison to dementia group, the number of 5-BrdU positive cells in the granule cell layer of normal control group was increased significantly .But the number of TUNEL positive cells was no difference from that of the dementia group'. The number of 5-BrdU positive cells and the number of TUNEL positive cells in the hilus of dementia group were no difference from that of the normal control group'.2. Part two: Methods:We develop the AD model through leisioning NBM by kainic acid . AD model rats received bFGF or saline by infusion into the cerebroventricular space every day after being lesioned.After 7 days bFGF infusion, 5-BrdUwas used to label the neural progenitors and the survival of newborn cells after bFGF infusion. TUNEL was used to label the apoptotic cell .Then we count the positive cells labeled for 5-BrdU or TUNEL in the dentate gyms. Results:Comparison to AD control group, the number of 5-BrdU positive cells in AD treatment group was increased significantly .But the number of TUNEL positive cells were no difference from that of the AD control group'. The number of 5-BrdU positive cells and the number of TUNEL positive cells were no difference from that of the nomal control group'. 3. Part three: Methods:We developed the AD model through leisioning NBM by kainic acid . AD model rats received bFGF(AD treatmemt group) or saline(AD control group) by infusion into the cerebroventricular space at 30 minutes ,1 day,3 days and 7days after being lesioned. The learning and memory abilities were measured by Y-maze test at 30 day after NBM lesioned; Follwing by AchE cytochemistry and Nissl's staining, calculation the density of the AchE fibre of hippocampus and forebrain. Results:Comparison to normal control group, learning and memory abilities of AD group were declined and the density of AchE fiber was decreased significantly (P<0.01) .Comparison to AD control group, learning and memory abilities of AD treatmemt group were improved and the density of AchE fiber was increased significantly(P<0.01). But all of the parametres of the treatmemt group did not reach the levels of that of the normal group' (P<0.01) .Conclusions:It is showed that:1.
