| Objective:Laboratory and clinical reports indicated that opiates have multiple effects on the parts of immune system. Macrophage are important immune cells and can present foreign antigents to T cells. Major histocompatibility complex(MHCⅡ) are necessary to finish presenting antigents and CD80 are the most important co-situmlatory molecules to active T cells. here, we assayed the influence of morphine dependence on expression of CD80 and MHCⅡfrom addictive mouse peritoneal macrophage.Material and methods: Forty-five BALB/c mouse were divided randomly into three groups, Every group include fifteen mice (three subgroup, n=5). According to EL-Kadi, mouse subcutaneous injection in increasing doses three times a day for seven days. Three morphine groups were injected morphine; three abstinent groups were injected nalonoxe before morphine; Three groups were injected saline as control group. On the 3rd,5th,7th day, peritoneal cells were harvested from three group mouse by peritoneal lavage with cold PBS. The peritoneal supernatant was washed, counted and adjusted to 1×106 cells/ml in RPMI-1640 medium supplemented with 10% heat-inactivated fetal calf serum. The cells were stained with phycoerythrin (PE)-anti-IAd or fluoresceinisothiocyanate (FITC) anti-B7-1 monoclonal antibody (Pharmingen) , the fluorescence was measured in a flow cytometer.Result:1. After injecting morphine for seven days, the model of morphine addiction was established. After naloxone administration, the morphine group mouse resulted in a remarked abstinence syndrome. The behavior score of abstinence syndrome were different from that of the control mouse(P<0.05). That means that the model of addiction on morphine had been developed.2. In the fifth day, result showed that in morphine-treated group the peritoneal cells increased the CD80 expression 45%(P<0.05). while the morphine plus naloxone group has increased also 33% but lower than morphine group (P<0.05). On the 7th day, these two groups remain the higher level.3. In the fifth day , the expression of MHCⅡ in morphine group and naloxone group has decreased 40.4% and 28% comparing the control group(P<0.05), and the morphine plus naloxone group decreased lowly than morphine group(P<0.05). IN the seventh day the morphine and naloxone group remain the lower level expression.Conclusion: our results show: ①Expression of CD80 increasing and MHCⅡ decreasing had significantly occured after addictive which likely was a important mechanism of depressing macrophage antigen-presentence function.②the drug is acting partially via classical opioid-receptor. In macrophage antigen-presentence produce , the both signs(MHCⅡand CD80) were important. |
