| [Objective] (1) To study HSP70 expression during limb ischemia reperfusion in rats. (2) To find out the influence and mechanism of heat stimulated preconditioning on limb ischemia reperfusion. (3) To establish new methods for the prevention and treatment of eschemia reperfusion injury.[Methods] (1) To establish a limb ischemia-reperfusion model and heat stimulated preconditioning model differencely in rats;(2) To record the change of anus temperature and heart rate. (3) To collect the serum of each period,measuring SOD activity with NET method,measuring the MDA contents with TBA method,measuring the contents of GOT and LDH with biochemistry analyzer;(4) To examine the HSP70 expression with Western-blot method;(5) To observe the histological structure changes of muscles tissue by light microscope.[Results] (1) At the time of Ih before ischema and 12h after ischemia reperfusion,there was no significant difference (P>0.05) in rats' anus temperature between IR group and NC group,while significant differences were recorded(P<0. 05) at Ih and 4h after ischemia reperfusion between the groups;Significant differences were recorded(P<0. 01) between HP+IR group and IR group in all thefour phases;(2) At Ih before ischemia,the contents of ALT and L-LDH in serum had no significant difference (P>0.05) among the three groups;At Ih,4h and 12h after ischemia reperfusion,the contents of ALT and L-LDH in HP+IR group and IR group were obviously higher (P< 0.01 ) than that in the NC group,while the contents in HP+IR group obviously lower (P<0. 01) than that in IR group. (3) At the time of Ih before ischemia,the content of MDA had no significant difference (P>0. 05) among the three groups,the contents of MDA in IR group were significantly higher (P< 0. 01 ) than that in NC group at Ih,4h and 12h after reperfusion,showing a continous increasing tendency after reperfusion;while at Ih,4h and 12h after reperfusion,the content of MDA in HP+IR group were obviously higher (P<0. 01) than that in NC group,lower than that in IR group,showing a continous increasing tendency following reperfusion. (4) At Ih before ischemia,the activity of SOD had no significant difference (P>0. 05) among IR group,NC group and HP+IR group,the activity of SOD in IR group were obviously lower (P< 0.01 ) than that in NC group at Ih,4h and 12h after reperfusion,and showing the continous decreasing tendency following reperfusion;while at Ih,4h and 12h after reperfusion,the content of SOD in HP+IR group were obviously lower (P<0. 01) than that in NC group,higher than that in IR group,showing the continous lowering tendency following reperfusion. (5) The expression quantity of HSP70 in NC group was little in skeleton muscle,it increased after ischemia reperfusion,showing a decreasing tendency after reperfusion;The expression of HSP70 increased obviously after heat stimulated preconditioning,showing a continous decreasing tendencyfollowing reperfusion. (6) In microscope (X 400):comparing with the IR group,the bright-dim zones in skeletal muscle cells were darity,membranes in small blood vessels integrated,and muscle cells swelled lightly in HP+IR group.[Conclusion] (1) The rats' anus temperature began to increase after heat stimulated preconditioning,and decreased following reperfusion,nevertheless,it was still higher than normal level;the heart rates of rats began to increase after heat stimulated preconditioning,and maintained at a higher level;(2) The contents of ALT and L-LDH in rats' serum after limb ischemia reperfusion decreased under heat stimulated precondition,it resembled a partial recovery of skeleton muscles;(3) The contents of ALT and L-LDH in rats' serum raised after limb ischemia reperfusion,they were sensitive indexes to detect the damage of muscle cells;(4) The content of MDA in rats' serum after limbs ischemia reperfusion decreased under heat stimulated precondition,while the SOD activity in rats' serum after limbs ischemia reperfusion was increased under heat stimulated precondition.
