| Intercellular adhesion molecue-1 (ICAM-1) is a critical adhesion molecule which mediates the adhesion between cells and cell base,or cells each other. It plays an important role in inflammatory reaction of the body .Recent research results manifested that ICAM-1 is expressed not only on renal glomerular endothelial cells(GEC) but also on tubular epithelial cells(TEC) at low levels.The expression of ICAM-1 is strengthened especially in tubulointerstitial injury.Many kinds of cytokines can regulate the expression of ICAM-1 on TEC,which may play pathogenic role in inflammatory reaction of tubulointerstitial nephropathy.The aim of this study was to detect the expression level of ICAM-1 on rat TEC(NRK52E cells) and the regulation of Tumor necrosis factor a (TNF a ) and Transforming growth factor (TGFB ) to expression of ICAM-1.Method:1. NRK52E cells was cultured and passaged .2. After stimulation by rhTNF a or rhTGF P (50ng/ml) for 1,4,12,24 hours,NRK52E cells was collected.Protein and total RNA was extracted from NRK52E cells.3 After stimulated by rhTNF a or rhTGF P (1 ng/ml,5 ng/ml,10 ng/ml,50ng/ml) for 4hours,NRK52E cells was collected.Protein and total RNA was extracted from NRK52E cellsAThe protein expression of ICAM-1 was measured by Western Blot. 5.The mRNA expression was determined by RT-PCR.Results:1.The protein and mRNA of ICAM-1 was expressed at low levels on nomoral TEC.2.After stimulated byrhTNF a,the expression of protein of ICAM-1 was strengthened gradually and reach the peak at 12hour. the expression of mRNA of ICAM-1 was strengthened gradually and reach the peak at 4hour.3. The expression of protein and mRNA of ICAM-1 was strengthened gradually with the rise of concentration of rhTNF a .4. After stimulated by rhTGF &,the expression of protein and mRNA of ICAM-1 was not strengthened.5.After prestimulated by rhTGF 3,the protein and mRNA of ICAM-1 was not induced by rhTNF a expect for 50ng/ml rhTNF a .Conclusions:ICAM-1 was expressed at low level on normal TEC. 3th protein and mRNA of ICAM-1 was upregulated by TNFa in dependent of concentration and time. ICAM-1 could not be upregulated by TGF P . TGF P inhibited the expression of ICAM-1 induced by TNF a . |
