Objective: To determine the suitable schemes of chemotherapy of 5-fluorouracil in clinic. Methods: 30 patients with colorectal adenocarcinoma were divided into A and B groups randomly. The adjuvant chemotherapy (AC) with leucovorin/5- fluorouracil (LV/5-FU) was carried out within two weeks after radical operation. Group A was treated by the four-week schedule of mainline of 5-FU, while group B by two-week schedule of 46 hour continuous infusion of 5-FU. Toxicity and phlebitis were observed during two weeks after AC each time. Data was analyzed statistically byx2-test. Results: Grade 1-2 stomatitis and grade 1-2 diarrhea and phlebitis were more in A than those in B. Conclusion: Although the dose of 5-FU was high in the two-week schedule , the two-week schedule was superior to the four-week schedule because of the low toxicity and the less phlebitis and the shortened time of being in hospital.
