| Objective: to make a quantitative analysis for the effects of decoction JinShuiLiuJunJian on basic biogenetic stereology parameters of bronchial mitochondria and nucleus in cilia-cell as well as goblet-cell of chronic bronchitis mice model.Search the mechanisms of decoction JinShuiLiuJunJian on chronic bronchitis.Methods: The chronic bronchitis mice model was made with sootiness, Slices of olefin and transmission electric-lens were made according to the general methods and observed with optics microscope and Transmission Electron Microscopy.The basic parameters were measured by Image Analysis Apparatus and biogenetic stereology,Results: In chronic bronchitis mice model group, there was an increase in Surface Number Density for bronchial goblet-cell (Na). A lot of mucus granules were on top of the goblet-cell. Quantitative analysis shows that Na of goblet-cell was 1.7 times more than that of in normal group( from 0.0028 ± 0.0007 to 0.0080± 0.0008,p<0.05 ). Mitochondria aggregated near the root of the cilia. The number decreased. Mitochondria were swollen. Cristae were shortened, dissolved and even disappeared. Stroma got thinner and electronic matrix density decreased .Comparing to the normal group , quantitative analysis indicates that Ratio of Surface to Volume for Mitochondria(Rsv) decreased 26.5% ( from 14.10± 1.61 to 10.36±0.93 ,p<0.05 ).Number Density(Nv) decreased 44.0% (from1.50± 0.57 to 0.84±0.30,p<0.05).Volume(vV) increased 92.9% ( from 0.14 ± 0.04 to 0.27± 0.09,p<0.05 ).But Volume Density (Vv) and Axial Ratio (Rab) had no changes. The volume of nucleus decreased obviously. Comparing to that of the normal group, Ratio of Surface to Volume of nucleus (Rsv) increased 94.6% (from 1.49±0.25 to 2.90±0.76 ,p<0.05 ).Volume Density(Vv) decreased 51.9% (from 0.54 ± 0.13 to 0.26 ± 0.06 ,p<0.05 ).But Surface Density (Sv) and Axial Ratio (Rab) had no changes. Comparing to the normal group, JinShuiLiuJunJian group had no changes (p>0.05),but were obviously different from the chronic bronchitis mice model group(p<0.05). Conclusions: Decoction JinShuiLiuJunJian can decrease the quantity of bronchial goblet-cell in chronic bronchitis mice model and can also improve the damages of the mitochondria and nucleus in cilia-cell, which is probably one of the important treatment mechanisms of decoction JinShuiLiuJunJian on chronic bronchitis. |
