| There is a tough problem that always puzzle neuroscientist. The problem is what we can do to improve the recovery of the injuried spine cord. Recently, great advances has been made on the research of spine cord injury. Many successful works are reported lately to enhance the axonal regeneration and functional restoration of the injuried spinal cord. Olfactory ensheathing cells (OECs) transplanting technique is the one of the bright achievements.OECs are glial cells in Olfactory system. OECs can supply suitable microenvironment for the axonal regeneration by secreting many neurotrophic facte^ cell matrix> nerve cell adhesive molecule. Another aspect OECs can form natural integration with central nervous system (CNS) . OECs can follow and parcel the regenerate axon, avoiding to contact with inhibition factor. OECs can lead axon regenerate to definiteTarget cell, forming synapse. Recently there are many successful research in OECs transplanting to improve axon regenerate. Especially there were recover of part motor function in spinal cord whole spoke model.There are two parts in this studies. The first part is the primary culture of human embryo OECs. We choose abortus of three to six months old as research object. In the research, we collected 35 specimen that all came from induction of labor with water bag. We successfully culture OECs from human embryo olfactory bulb by using primary culture. During the specific time periods,we appraised cell and calculate the proportion of positive by immunochemistry Staining. We find the period after primary culture fifteenth day is the best period for transplant operation.In the period, the proportion of positive of OECs is highest and can arrive 90%. We have fumbled about a new cell culture method. First we draw the materials from specimen, then put in culture medium to maintain on 4C .After consummatd every conditions,we can cultivate. Conclusion:our research result verify the feasibility of culturing OECs from embryo, provide a condition for restoring spinal cord . injury by using OECs.The second part is to study the effect of Basic f ibroblast growth factor (bFGF) on rat' s olfactory ensheathing cells growth in vitro. OECs were separated from 2.5 months old adult rat' s olfactory bulb and inoculated in 24-hole culture plate, and primary culture was proceeded. At the 7th day being cultured, differentconcentrations of bFGF (20ng/ml, 10ng/ml, 5ng/ml) were added into each hole. After 72 hours, the cells were counted under microscope. Cellproliferation rates were acquired from comparing the cells counting before and after added bFGF. The cellproliferation rate was highest when added into 10ng/ml bFGF, compared with the control group , 20 ng/ml and 5 ng/ml group,the difference was significant. The outcome was analysised statistically. At the another hand, the proportion of positive of OECs in every group are not difference.Conclusion: we can think bFGF can facilitate OECs growth, and depend on its concentration without influencing the proportion of positive of OECs. It will provide experimental data for spinal injury therapy with OECs transplantion. |
