Weitongxin is composed of 6 kinds of Chinese herbs: Radix Et Rhizoma Rhei, Radix Aucklandiae, Fructus Gleditsiae Abnormalis, Semen Pharbitidis Album, Semen Pharbitidis Niger and Radix Glycyrrhizae . This prescription is the recipe of proven efficacy with the purpose of curing gastric cavity ache. Owing to its remarkable effect, the prescription is well worth further studying and developing . This paper is about the extraction process , quality assessment and research of pharmacodynamics and toxicology. TLC and HPLC specification of Weitongxin provide its quality assessment.The optimal process was studied by orthogonal design with yield of the rhein, emodin content and extract weight as indexes. Three factors were selected in this experiment, including the concentration of solvent ethanol, the duration of extraction and times of extraction. The result showed that the optimal extraction process was : 90%ethanol, extracting for2 times, 4hours each time.Radix Et Rhizoma Rhei, Radix Aucklandiae, Fructus Gleditsiae Abnormalis, Semen Pharbitidis and Radis Glycyrrhizae in Weitongxin were identified by TLC with the posts clear and reproducible. Rhein, emodin, chrysophanol and echinocystic acid in Weitongxin were determined by HPLC. They had good linearity within the range of: 0.03ug~0.48ng (r=0.9996) , 0.025ug~0.40ug (r=0.9997) , 0.025 g~0.40 g (r=0.9996) , 0.15 g~ 2.4[ag (r=0.9998) respectively. The average recoveries of them were 97.2% ( RSD=1.6% ) , 96.4% ( RSD=2.1% ) , 95.0% ( RSD=1.6% ) , 97.8%ABSTRACT(RSD=1.7%) respectively.The results of pharmacodynamics and toxicology show that: Weitongxin had marked antiulcer effect on several kinds of ulcer model tests which accelerated ulcer healing in rats with mucosal lesions induced by HAC when administered orally. Weitongxin significantly increased the contractile activity of isolated gastric muscle strips in rats, and increased the qercent of charcoal transit in mice. Weitongxin significantly decreased the writhing frepuency induced by acetic acid in mice. Acute toxicity of Weitongxin show that: its toxiccity is so little that nowas detected. |
