| objective: Judge the effects of different concentrations by NaF resorptive activity of isolated osteoclasts.lt has been developed a method of estoclast culture and observation of its apoptosis in vitro in order to unveil pathology variety of jaw in toxic fluorosis.Method: Osteoclasts were obtained from the 24-h newborn Japanese rabbit. We had analyzed the effects of NaF on the activity of isolated rabbit osteoclasts cultured on thin slices of devitalized compact bovine bone. Osteoclastic resorption was quantitated by counting the number of resorption lacunae and measuring their surface area and their depth by using scanning electron microscopy.Results: Our results show that NaF in concentrations of NaF(10-4 10-5和 10-6mol/L) decreased the number of resorption lacunae made by individual osteoclasts and decreased the resorbed area per osteoclast. But at low dose of NaF the phenomenon (10-7mol/L) was not obvious.Conclusions: It has concluded of rabbit osteoclast in vitro were inhibited by NaF(10-4, 10-5 和 10-6mol/L) in dose-time dependent manner. As compared with control group, low dose of NaF (10-7mol/L) has been not effect osteoclast. |
