Objective To explore if the sequential culture technique more suitable for in vitro development of cultured 2 - 4-cell mouse embryos than classical single culture technique under our laboratorial situation. Method(s) 2-4-cell mouse embryos collected from the oviducts of the superovulated Kunming species white mice were cultured in microdrops, and the embryos were distributed to the single culture medium group and sequential culture medium group randomized. The percentages of embryos developed to 8-cell embryos stage, morula stage and blastocyst stage were used as evaluation criteria. Result(s) There are significantly higher proportion of 2-4-cell mouse embryos reached the 8-cell stage (since 46 out of 62, 74.19%, p=0.022), the morula stage (44 out of 62, 70.97%, p=0.041) and the blastocyst stage (40 out of 62, 64.52%, p=0.019) in sequential culture medium group as compared those in conventional single culture medium group (47 out of 83, 56.63%, 46 out of 83, 55.42%, 38 out of 83, 45.78%). Conclusion(s) (1)Sequential culture system is more suitable for in vitro development of mouse embryos, and could obtain higher quality embryos. Sequential culture offered a safe and possible means of producing blastocysts. (2) It is feasible to do sequential culture for human embryos under our laboratorial situation.
