PrefaceOsteoporosis was put forward by pommer in 1885 , as a systemic skeletal disease characterized by low Lone mass and niicrcarchitectural deterioration of Done tissue, with a consoquent increase in bone fragility and susceptibility to fracture risk, with the aged structure of population , osteoporosis has become more and more serious health problem in the whole world . Nowadays it is considered that osteoporosis can be caused by many factors such as hormone control, nutrition stale, physical factor and immunity function.Nitric oxide ( NO) plays a important role in nerve conduction , blood vessel diastole and immunity defence which is closely related with occur of many illness . in recent years , biologists found NO and NOS play important roles in association and promotion of pathophysiol-ogy course in osteoporosis .Researches indicate that NO , NOS are closely related with oste-oblast ( OB) and osteoclast ( OC) in bone . relationship between OC function and NO was first put forward by Maclntyre . study has proved that NO maintains the control of bone absorption through restrain quantity and activity of OC . NOS' restrain agent changes metabolic activity of bone through improving OC absorption to bone . Rianchofound OB be considered a lateral secretory factor to restrain bone absorption through generating NO . NOS with lower lever composed lower density NO so as to promote bone absorption . It indicated NO maybe played important role during the signal conduction course between OB and OC . exploring the relationship between NO , NOS and osteoporosis is very profound significance in order to find method of postponing or treating osteoporosis . but research of NO , NOS on osteoporosis just be opened in recently years , seldom relative report has been reported in china , up to the present the understand is still premilinary in NO , NOS to bone absorption controlling mechanism and its function of occurrence courses in osteoporosis . It need to deepen exploration . this experiment reveals NO , NOS important function in occurrence of osteoporosis.The reason of postmenopausal ( PMO) osteoporosis is not clear yet , so the researches on illness mechanism , prevention drugs function mechanism , drug effort and safety must apply animal test . female adult ovariectomy ( OVX ) rats similarity to menopausal woman in lowing bone mass , decreasing bone density , larging bone transformation . It was decided in 1980 that female adult OVX rats were generally recognized standard model of original osteoporosis . statistical result was a standard whether model is successful or not .This experiment firstly builds rats osteoporosis model and secondly measures the change of serum NO , NOS in two group of rats . analyzing the relativity between NO , NOS and osteoporosis . the aim is to provide experimental evidence for development of osteoporosis special treatment .Materials and methods1. animal and group, six months, adult female SD rats weight 250 25 g , total 30 . are devided random two groups; A: OVX rats model group of osteoporosis , B : control group.2. build osteoporosis model . rats are anesthetized in abdomen with 2.5% thiopental sodium according the rate weight 0. Iml/lOOg. A group rats are resected tv;o sides ovary. B group are resected omen-turn majus . test time is 13 weeks.3. measure rats bone mineral density ( BMO)13 weeks later since model, measuring two group of rats lumbar verebra BMD.4. collection of specimen. 13 weeks later since model , blood collecting from superior mesenteric artery are devided serum >0. 5ml, and placed rapidly into the refrigerator of twenty degrees beneath zero until die measurement of test.5. statistical analysis, use SAS software to analyse the results of the experiment date ,the level of significance is P <0. 05.Results1.
