At present, gastric acid has been affirmed to be one of the mostly pathopoiesia elements leads to peptic ulcer and other disordered diseases with gastric acid concerned ( such as reflux esophagitis). Reduce gastric acid secretion is the most frequently - used means. In recent years, medicine research tends to selectively restrain gastric acid. So proton pump inhibitor is widely used in kinds of disordered diseases with gastric acid concerned. Chinese traditional medicine Scopaparia dulcis, is used to cure cough, children hives, eczema, hypertension and erysipelas etc. In 1988, Hayashi reported that dry Scopaparia dulcis extracted by ethanol, the extractive is checked up, its main ingredient is diterpene compound scopadulcic acid B (SA - B). Kawasaki investigated it and found that SA - B can inhibit gastric H+/K+ -ATPase activity of hog(IC50 = 20 -30uM) and proton transferred into gastric vesical ( proton englobed) , which points out that SA - B can inhibit gastric H+/K+ -ATPase activity. Before long, Shinji Asano thoroughly studied the inhibition of gastric H+/K+ -ATPase by SA -B and its debenzoyl derivative, diacetyl scopadul ( DAS). He found that, both compounds dose - dependently inhibit the gastric H+/K+ -ATPase action without any effect on the Na +/K+ - ATPase activity. And the inhibitory action caused by SA - B and DAS recovered when diluted, which means the inhibition is reversible. This articleaims to study the Scopaparia dulcis' extractive from ethanol, role in gastric H+/K+ - ATPase activity by animal experiment, and observe its therapeutical effect on gastric ulcer. We want to thorough research its medicinal value, hope to develop a new proton pump inhibitor, it may be helpful to the clinical experiment in the future.Object and MethodTwo Japan white rabbits were selected to observe the diversification of gastric PH titer in 24 hours before and after fed medicine for a week, so as to estimate medicine, s inhibitory effect on gastric acid secretion. 30 wistar big white rats were divided to two groups; experimental group arid compare group, there were 15 in each group. Ah1 rats were made to be gastric ulcer model by acetic acid inustion. The rats in experimental group were fed with Scopaparia dulcis' extractive from ethanol , Ig/kg. d, the others were not fed. All rats were paunched and their stomach were obtained, the volume of ulcer was measured, to observe the contrast of ulcer volume between two groups, so as to estimate the curative effect of Scopaparia dulcis' extractive from ethanol on rats "gastric ulcer. Then removed the sinuses ventriculi , scrape the gastric mucosa, and measured the gastric H+/ K+ -ATPase activity, to appraise the effect of Scopaparia dulcis'extractive from ethanol on rat's gastric H+/K+ -ATPase activity.ResultsScopaparia dulcis'extractive from ethanol can inhibit the rabbit , s gastric acid secretion.Scopaparia dulcis' extractive from ethanol can also promote the cicatrize of rats' gastric ulcer. The volume of gastric ulcer in experimental group is less than that in compare group. There is difference in statistics.The gastric H+/K+ - ATPase activity of rats in experimental group is less than that in compare group , this declares that the Scopaparia dulcis'extractive from ethanol can restrain the H+/K+ -ATPase activity.DiscussionAt present, gastric acid has been affirmed to be one of the mostly pathopoiesia elements leads to peptic ulcer.
