The development of precutanous transluminal angioplasty provide a new way to treat the vessel constriction or obstruction. It's trauma is smaller and it's effect is confirmed, medical personal and patient were attracted. Recent randomized studies show 30% ~ 50% angiographic restenosis rates after angioplasty, so restenosis become the major limitation of this procedure. Until now, we still don't know the mechanism of restenosis and have no program to reselve it. There were many experiments about how estrogen protect cardiovascular functions, some scholar also found that estrogen can prevent the restenosis after angioplasty, but there is argument about estrogen can prevent restenosis in males. There is no related articles about androgen's function in restenosis. In this study, we observed the developmental changes of adventitia in restenosis, and investigated the relationship between sex hormone and restenosis after angioplasty.Part I The Effect of Castrating on Restenosis of Male Rats afterAngioplastyPurpose: To investigate the effect of physical androgen on restenosis and to discuss the mechnism by observing the changes of adventitia. Material and Methods: 1) To create the castrated models of male rats.2) To create the models of restenosis of left common carotid artery. 3)To observe the changes of adventitia cells in restenosis process throng labeling them by BrDU.4)To get specimen at 3rd days, 7th days, 14th days and 28th days. And to observe the condition of restenosis using hematoxylin and eosin staning , immunohistochemistry and electronic microscopy.Results 1) Adventitia activated after 3days, proliferating and changing phenotype; mostly proliferated after 7 days and migrated towards to middle layer; Proliferating adventitia decreased after 14 days, some adventitia cells migrating to neointimal; almost none be seen after 28days.2) Adventitial proliferating index of castrated group was larger than that of non-castrated group in Sdays ^ 7 days -. 14days after angioplasty. 3) the areas of adventitia and neointimal and the degree of restenosis are clearly larger in castrated rats, while caval areas are smaller than non-castrated group in 7 days > 14 days^ 2 Sdays after angioplasty. Conclusion 1 Activated adventitial contribute for restenosis after PTA by proliferating and migrating. 2 Androgen in physiology rat can relieve restenosis perhepers through interventing the activating of adventitia.Part II The Experimental Study of Inhibition of Restenosis afterAngioplasty by Smearing Estrogen on AdventitiaPurpose: To in investigate the effect of estrogen on restenosis after angioplasty by smearing the mixture of estrogen and pluronic gelatine, and to discuss the mechanism.Materials and Methods: 1) To create the castrated models of female rats.2) To create the models of restenosis of left common carotid artery. 3) 40 male rats were divided into the control group and the test group, 60 female rats into castrated group, non-castrated group and castrated +E group'. To get specimen in 3 days, 7 days, 14 days and 2 8 days after angioplasty and to observe the condition of restenosis. Results: 1) Adventitial proliferating index of male test group was larger than thatof the contrast group in 3 days,7 days, 14 days after angioplasty. The areas of adventitia and neointimal and the degree of restenosis are clearly smaller in test group, while caval areas are larger than the contrast group. 2) Adventitial proliferating index of non-castrated group and castrated+E group was smaller than castrated group, while no difference between non-castrated group and castrated+E group in 3 days, 7 days, 14 days after angioplasty. The areas of adventitia and neointimal and the degree of restenosis are clearly smaller in non-castrated group and castrated+E group, while caval areas are larger than the castrated group, while no difference between non-castrated group and castrated+E group.
