| Objective To study the effect of derivative of chitin on fracture healing and further investigate its therapetic mechanism.Method Eighty healthy Newzealand rabbits were made into models with 3mm bone defect in middle of segment of radius. They were randomly devided into 4 groups. They were given normal saline, bonesetting tablate, N-acely-glucosamine and chitowoligosaccha-ride daily through stomach tube respectively. Each group was divided into four intervals of 9,17,30and42 days postoperatively. X-ray, histological study were used to observe the healing. Immunochemistry technical was applied to observe the expression of BMP and TGF-J3 proteins.Result Fracture healing reflected by X-ray and histological stain showed significant difference(P<0. 05) between groups fed with derivative of chitin and controls treated with normal saline and bonesetting tablate 9 and 17 days after operation. The difference was also significantly(P<0. 01) 30 and 42 days following operation. Immunochemistry study manifested significantly increase of BMP and TGF-β levels in fracture ends in groups fed with derivative of chitin in different phases than controls.Conclusion The derivative of chitin can promote osteoblast activity and collagen formation during fracture healing,increase the expression of BMP and TGF-β in the healing process and accerlerate fracture healing. |
