ObjectiveTo study the feasibility of growth, expansion, differentiation and labeling invitro of the bone marrow stromal cells (BMSCs) of the rhesus monkey.Materials and methodsThe BMSCs were isolated from the adult rhesus monkey, cultured andinduced with some cytokines and special medium confected in our lab (patentnumber:02134314.4), and labelled with BrdU. Identification was performedwith immunochemichemistry method.ResultsThe BMSCs could proliferate and differentiate into neural stem cells whencultured in vitro. These cells might grow rapidly and further differentiatedinto neuron-like cells and astrocyte-like cells. Cells clone proliferated fromstem cells could express Nestin or Vimentin antigen, while the differentiatedcells might express GFAP and NSE antigen respectively. It was alsoconfirmed that the cells differentiated neural stem cells from the BMSCscould be labelled with BrdU. In this study, no special effects RA, bFGF and EGF on the induction and differentiation of BMSCs derived neural stem cells were remarkably found. ConclusionThe BMSCs from rhesus monkey possess the abilities of self-renew and differentiation. They might differentiate into the Nestin-positive and Vimentin-positive neural stem cells, and further into the neuron-like and astrocyte-like cells which expressed GFAP or NSE. As a convenient and efficient marker, BrdU could be used as an optional tracer for cells transplantation. As available easily, the BMSCs could be considerd as the ideal seed cells of the neural stem cells.
