Objective:To study the expression and the effect of TGF_β and VEGF on the repairing bone defects and the combination of musculoperiosteal flap,bone marron and spongy bone granules in transplanted to repair the bone defects. Method: Segmental bone defects for 2.0cm were created in the mid_upper of radial shaft of adult rabbits. The defects were filled with combination of musculoperiosteal flap,bone marron and spongy bone granules. The samples are taken 3d. 1,2,4,6,8 weeks postoperatively. Studied by optical microscope., immunohistochemistry. Compared with each other to decide the effect of repairing . The healing of bone defects are observed carefullyin each periods. Results: It was found that the expression of TGF_β and VEGF changed in different periods. The cell in the cambial layer of the periostum showed low or negative signal in the immediate injury response period. TGF_β and VEGF were expressed in many kinds of cells in the bone defects site during 3d to 6 week after operation. The high expression of TGF_β and VEGF were detected by the 4 week.. Decreased signals for TGF_β and VEGF were observed by 6 week. The experimental group had a better expression than control group (P<0.05). . Conclusion: combination of musculoperiosteal flap,bone marron and spongy bone granules can repair the long bone defects. TGF_βand VEGF are key factor in healing bone defects. : combination transplant is benefit to repair bone defects. Considering the satisfactory osteogenesis, osteoconduction and osteoinduction seen with this method.
