Objective:On the base of FFDS being used to treat cardiac disease, we research the acting mechanism of the main component-Sal B in herbs. We would explain the passway of Sal B in injure of hypoxia on CMEC and CM.Method:Using the technique of CMEC and CM culture, we established the model of hypoxia preconditioning. Basing of the mechanisms, we use the rate of live cell (MTT), lactate dehydrogenase (LDH), superoxide dismutase (SOD), malondialdehyde (MDA) and ICAM-1 act as the evaluating criterion, to observe the different effects in the different conduction passway.Result: 1. Under the hypoxic culturing condition, the livability of CMEC and CM effected by Sal B can be enhanced. When the cell was cultured by Sal B in short time, the effect is significant. 2. Using the method of ELISA, we can detect that the expression of ICAM-1 on CMEC is increased after hypoxia and reoxygenation. 3. The expression of ICAM-1 on CMEC can be reduced by hypoxia preconditioning. So dose the Sal B. 4. Chelerthrin can not block the effect of hypoxia preconditioning in the expression of ICAM-1 on CMEC. 5. Under the hypoxic culturing condition, SalB can play the role of hypoxia preconditioning. Sal B can enhanced the livability of CMEC and reduce the release of LDH significantly. 6. When we used chelerthrine to affect CM, the effect of hypoxia preconditioning was disappear. The phenomenon was happened that the livability of CMEC was reduced and the release of LDH was increased. But, when we used Sal B, there would happen opposite phenomenon. 7. Glibenclamide could block the effect of hypoxia preconditioning on CM too. However, Sal B can enhance the activities of SOD and the livability of CM. It can reduce the content of MDA and the release of LDH.Conclusion: 1. Under the hypoxic culturing condition, Sal B can bring about theeffect of protection on CMEC and CM in short time. 2. Sal B can reduce the expression of ICAM-1 on CMEC. 3. PKC-K+ATP conduction way participate the protective effect to CMEC and CM. Sal B can bring about the protective effect to CMEC and CM through the PKC-K+ATP conduction way. However, the target is still not un-definition. It may be in K+ATP, because the K+ATP locate the downstream of PKC.
