Piezoelectric biosensor using quartz crystal as transducer is a sensor that can translates the detecting signal into electric signal. Genechip also named DNA "microarray" is an important kind of biochip. We can achieve this kind of biochip by immobilizing a great many of probes on a weeny substrate or designing micro-cell, to realize the exact, speedy detection of compounds such as protein, nucleic acid, cell and other biological compounds. It also can scan a great deal of information. In the thesis, we analyze the human immunodeficiency virus's by piezoelectric biochip.In this thesis, we research the optimum condition of immobilizing single-chain DNA on the surface of gold-filled quartz crystal. By experiments, we ascertain that under the condition of pH8.1 and ion intensity 0.016, the oligonucleotide probes used in the experiment can be immobilized satisfactorily. At the same time, we also study the reactive condition including immobilizing time, hybridize solution, hybridizing time and so on. We founded a piezoelectric biochip of HIV-1.In the thesis, we immobilize the oligonucleotide on the surfaces of gold-filled quartz crystal by the self-assembled technique. According to the principle of "base pair mutual-reinforce", the nucleic acid hybridizes idiosyncrasily. Then we analyze the nucleic acid of HIV-1 using the piezoelectric biochip real-time apparatus AGA2002. Being the initial stage of clinical application, it was mainly used in the gene analysis of the nucleic acid of HIV-1 (RNA).About the HIV-1, on the condition of no PCR process, we used the AGA2002 to detect the nucleic acid directly after it was purified from the serum. We had the experiment in the condition of blind, that is to say we knew no information about the specimen. Comparing the detection result of AGA2002 with Bayer 3.0 of HIV-1 apparatus, the result indicated: the accordance result of negative and positive is very good; the negative is 100% and the positive is 94.9%. Using the HBV and healthy human blood as antitheses to detect the idiosyncrasy of the technique, the result is satisfying. This technique can fit the qualitative analysis of HIV-1. In this thesis, according to the condition of no polymerization chain reaction process we deduced two response models that could be not only applied in clinical detection successfully, but also applied in the studies of hybridizing kinetics and thermodynamics. We also use the equity model analyze the result of HIV-1 detection.On the condition of not having polymerization chain reaction, we got half-quantitative analysis result. Finally, the thesis discussed the application foreground of piezoelectric biosensors for nucleic acid analysis.
