| Objective: Day 1-8 of mouse pregnancy after fertilization is a period during which embryo develops,enters uterus and recognizes,adheres with endometrium luminal epithe lium,implants in the endometrial stroma,until placental circulation is established. Embryo implantation is a key step and a complex process.Many factors are involved in the process,such as hormone, cytokines,adhesive molecules,cell surface oligosaccharide, extracellular matrix,matrix metallopreinases,which consist of a network to control this process meticulously.Researches in recent years have showed cell surface stage specific oligosaccharides play an important role in this process.Single fucosylated Lex is involved in the compaction of cleavage-mass. Double fucosylated Ley is closely related to implantation.Masking Ley on the surface of endometrium or embryo with its specific antibody, implantation is significantly inhibited.Researches have also demonstrated interaction between receptive endometrium and invasive blastocyst only occur at specific stage(So called implantation window). Establishment of receptive endometrium is controlled by hormone and many factors are also involved.MMP-2 is an enzyme that is widely involved in extracellular matrix remodeling.MMP-9 is the enzyme participating in trophoblast cell invasion. Masking MMP-9 with its antibody can inhibit trophoblast cell invasion in co-culture system. TGF β 1 is involved in the creating of receptive endometrium through promoting the synthesis of some proteins,decreasing proteolytic activity and regulating integrin synthesis.This dissertation quest function of Ley ,Lex,MMP-2, MMP-9 and TGF β 1 during implantation through locating their expression,so as to provide new ideas for clinical assistant reproduction and antiimplantation contraception.Method: Blue dye solution was injected intravenously(tail vein)5 min before mice were killed to identify implantation segments on days 5 and 6,and then tissue section were investigated by immunohistochemical method.Result: 1.During 1-4 days of early pregnancy, Ley was detected in the luminal and glandular epithelium.On day 4,before the onset of implantation, the greatest staining was noted.After implanting ,on day 5, Ley remained in the luminal epithelium, on day 6 staining became weak at implantation site.This pattern of staining remained the same in inter-implantation areas on days 5-8.2.During 1-4 days of early pregnancy,expression of Lex gradually became weak in the luminal epithelium,but it gradually became strong in the glandular epithelium and the staining was the greatest on day 3.After implanting, on day 5 Le" was not expressed at the conceptus-maternal interface, except luminal epithelium without the conceptus-maternal interface. In inter-implantation areas expression gradually became weak in the glandular epithelium,this pattern of staining remained the same in the luminal epithelium.3.Before implanting,MMP-2 were detected in the luminal and glandular epithelium,both sites had the greatest staining on day 3. After implanting,on day 5 staining became the greatest in luminal epithelium of implantation site.Although immunoreactive MMP-2 remained in the stroma in implantation segments,staining was seen only in the layer of non-differentiated stromal cells adjacent to the myometrium on days 6 and 7.On day 8 staining was seen in whole stroma,predominantly in mesometrial pole. In inter-implantation areas expression did not change in the luminal and glandular epithelium and staining wasnot seen in the stroma.4.Before implanting,MMP-9 expression are identical in the luminal and glandular epithelium.On day 4,before the onset of implantation both had the greatest staining and staining gradually became stronger in the stroma. After implanting, on days 5 and 8, MMP-9 were detected in whole stroma of implantation segments, staining was located mainly outside the decidualized zone on days 6 and 7,in the layer of non-differentiated stromal cells adjacent to... |
